Abstract

This study presents two experimental series in mice aimed at determining optimal dosing strategies for cyclophosphamide and its impact on blood parameters when combined with vitamin B12. In the first series, two groups of mice received cumulative cyclophosphamide injections via tail vein over 25 days. Group I received escalating doses from 100 to 300 mg/day equivalent; Group II started at higher fixed doses (200–400 mg/day). Mortality was significantly higher in Group II, demonstrating better tolerance in gradually increased dosing schedules. In the second series, cyclophosphamide and nitromine were administered with or without vitamin B12. Retroorbital blood sampling was used to monitor leukocyte and erythrocyte counts. No conclusive hematological protection by B12 was observed under the tested conditions, though methodological limitations (e.g., dosing window) were noted. The author emphasizes the need for further trials in expanded protocol scopes.

Cyclophosphamid'e Karşı Farelerin Tahhümmülü ve Cyclophosphamid ile Nitrominin B12 ile Tatbikine Ait Deneyler

Abstract

Bu çalışma, farelerde iki farklı deney serisi ile siklofosfamid kemoterapi ajanına karşı tolerans ve hematolojik etkiyi B12 vitamini ile birlikte değerlendirmiştir. İlk deneyde iki grup fareye 25 gün boyunca kuyruk toplardamarından kademeli (Grup I: 100–300 mg/gün) ve sabit (Grup II: 200–400 mg/gün) dozlarda siklofosfamid uygulanmıştır. Grup II’de mortalite çok daha yüksek olup, ilaca karşı daha iyi tahammülün kademeli doz artışıyla sağlandığı görülmüştür. İkinci deney dizisinde, siklofosfamid ve nitromin B12 vitamini eşliğinde ya da yalnız uygulanarak farelerin retroorbital veninden alınan kan örnekleriyle lökosit ve eritrosit sayımları yapılmıştır. B12’nin hematolojik yan etkilere karşı koruyucu etkisi bu koşullar altında belirgin şekilde saptanamamıştır. Yazar, doz ve süre parametrelerinin genişletildiği yeni deneylerin gerekliliğini vurgulamıştır.
