Nefron'da Asid Fosfatazın Elektron Mikroskopik Lokalizasyonu

Abstract

Asid fosfataz'ın elektron mikroskopik lokalizasyonunu göstermek için, kobay böbreğinden alınan taze korteks parçalarından cryostat mikrotomu ile hazırlanmış 40 mikron kalınlığındaki kesitler üzerinde, kısa formaldehid fosfat fiksasyonundan sonra asid fosfataz reaksiyonu tatbik edildi, sonra bu kesitler elektron mikroskopik teknik materyeli olarak kullanıldı. Asid fosfataz aktivitesinin mevcudiyetini gösteren siyah kurşun sülfür'den ibaret pozitif reaksiyon ürünü, tubulus proximalis epitel hücrelerinin daha ziyade apikal yarımında, dens (yoğun) granulalar ve rezorpsiyon (sindirim) vakuolleri üzerinde, ayrıca bu çalışmada kullanılan materyelin (kobay) özelliği olarak da pinocytose vezikülleri ile mikrovillusların dibinden cytoplasma'ya uzanan tubuler envaginasyonlarda görüldü. Glomerulusda ise, epitelyal hücrelerin periferik cytoplasma'sında yine dens granulalar ve rezorpsiyon vakuollerinde çok daha az miktarda bulundu. Glomerulus rezorpsiyon vakuollerinin pozitif reaksiyonu hakkında literatür bilgisine rastlanmadı. Nefronun diğer bölümlerinde asid fosfataz reaksiyonu negatif oldu.

Electron Microscopic Localization of Acid Phosphatase in the Nephron

Abstract

To demonstrate the electron microscopic localization of acid phosphatase, the reaction was applied on 40-micron thick cryostat sections prepared from fresh cortex pieces of guinea pig kidney, after a short formaldehyde phosphate fixation; these sections were then used as material for electron microscopic technique. The positive reaction product, consisting of black lead sulfide indicating the presence of acid phosphatase activity, was observed in the epithelial cells of the proximal tubule, mostly in the apical half, on dense granules and resorption (digestion) vacuoles, and also, as a characteristic of the material (guinea pig) used in this study, in pinocytotic vesicles and tubular invaginations extending from the base of the microvilli into the cytoplasm. In the glomerulus, it was found in much smaller amounts in the peripheral cytoplasm of epithelial cells, again in dense granules and resorption vacuoles. No information was found in the literature regarding the positive reaction of glomerulus resorption vacuoles. The acid phosphatase reaction was negative in other parts of the nephron.
