Diploit Ve Tetraploit Asma Genotiplerinde Polipolidi Doğrulama Yöntemlerinin Testi

Abstract

Poliploidizasyon yüksek bitkilerde evrimi yönlendiren ana faktörlerden biri olarak kabul edilmektedir. Poliploidizasyonun bazı bitki fenotiplerini değiştirdiği ve temel özelliklerin çoğuna dokunmadan bırakması nedeniyle üzüm çeşitlerinin kalite, verim veya çevresel adaptasyon gibi belirli özelliklerin geliştirilmesinde faydalı olması beklenmektedir. Bu çalışmada iki diploit Ekşi Kara ve Gök Üzüm çeşitleri ile iki tetraploit Kyoho ve Heuk Boseok üzüm çeşitlerinin ploidi düzeylerinin tespitinde ön eleme verileri için kullanılan stoma özellikleri (yoğunluk, uzunluk, genişlik), kloroplast sayıları (adet stoma), yaprak klorofil içerikleri (SPAD değeri) ve yaprak kalınlıkları (m) karşılaştırılmıştır. Bitkisel materyal örnekleri serada vejetatif olarak çoğaltılmış 35 litrelik saksı içerisindeki 2 yaşlı bitkilerden alınmıştır. Sitolojik çalışmada, denemede kullanılan çeşitlerin tümünde Flow Sitometri (FC) analizi yapılarak veriler karşılaştırılmıştır. Diploit üzüm çeşitlerinin stoma sayıları tetraploit çeşitlerinkinden önemli ölçüde fazla, stomaları daha yoğun, stoma uzunluk ve genişlik değerleri daha düşüktür. Tetraploit çeşitlerin yaprak klorofil içerikleri ile yaprak kalınlıkları diploit çeşitlerinkinden önemli ölçüde daha fazla, kloroplast sayıları ise diploit çeşitlerin yaklaşık iki katıdır. FC analizlerinde tetraploit çeşitlerin pik seviyesi diploitlerin iki katıdır. Yapılan incelemeler sonucunda tetraploit çeşitler ile diploit çeşitler arasındaki farklar tüm parametrelerde önemlidir. Stoma, kloroplast, klorofil ve yaprak kalınlığı parametrelerinin poliploit bitki ıslahı çalışmalarında, çok sayıda örnekle çalışırken poliploit bitkilerin tespitinde araştırıcılara zaman kazandırmaktadır. Ploidi düzeylerini doğrulama için FC analizi gerekli görülmektedir.

Testing Of Polypolydy Confirmation Methods In Diploit And Tetraploit Grapevine Genotypes

Abstract

Polyploidization is accepted as one of the main factors driving evolution in higher plants. Because polyploidization alters some plant phenotypes and leaves many of the essential traits untouched, grape varieties are expected to be beneficial in improving certain traits such as quality, yield or environmental adaptation. Stomatal characteristics (density, length, width), chloroplast numbers (number stomata), leaf chlorophyll contents (SPAD value) and leaf thicknesses (m) are the pre-screening data in determining the ploidy levels and in this study, two diploid Ekşi Kara and Gök Üzüm varieties and two tetraploid Kyoho and Heuk Boseok grape varieties were compared to these methods. Plant materials samples were taken from 2-year-old plants in 35-liter pots, which were propagated vegetatively in the greenhouse. In the cytological study, Flow Cytometry (FC) analysis was performed on all cultivars used in the experiment and the data were compared. The number of stomata of diploid grape varieties is significantly higher than that of tetraploid varieties, the stomata are denser, and the stomatal length and width values are lower. Leaf chlorophyll contents and leaf thicknesses of tetraploid cultivars are significantly higher than those of diploid cultivars, and chloroplast numbers are approximately twice that of diploid cultivars. In FC analyses, the peak level of tetraploid cultivars is twice that of diploids. As a result of the examinations, the differences between tetraploid varieties and diploid varieties are important in all parameters. Stoma, chloroplast, chlorophyll, and leaf thickness parameters save time for researchers in the detection of polyploid plants while working with many samples in polyploid plant breeding studies. FC analysis is considered necessary to confirm ploidy levels.
