Ceviz (Juglans regia L.) Çeşitlerinin RAPD Tekniği İle Genetik Tanımlanması

Abstract
Bu çalışmada RAPD markörleri ile 26 adet ceviz çeşit ve seleksiyonunun karakterizasyonu yapılmış ve aralarındaki genetik ilişkiler belirlenmiştir. DNA saflık değerleri 260/280:1.87 ve 260/230:2.0 olarak tespit edilmiştir. DNA miktarı ortalama 44.6 ng/µl olup 12.3-84.1 ng/µl arasında değişmiştir. 44 Operon primeri tekrarlanabilir ve net 301 adet polimorfik bant üretmiştir. Primerler 2-17 adet arasında ortalama 6.85 bant oluşturmuştur. 18 çeşitte tek bant, 8 çeşitte ise 2-3 bant kombinasyonu ile çeşit tanımlaması yapılmıştır. Genotipler arasındaki Dice genetik benzerlik indisi 0.42 ile 0.94 arasında değişmiştir. Birbirine en yakın genotipler Kaplan-86 ve Y1 olurken en farklı genotipler 65/7 ve Hartley bulunmuştur. UPGMA dendogramı, genotipleri 3 farklı grup altında toplamıştır. DNA kaliteli olduğu ve reaksiyon koşuları optimize edildiği sürece RAPD tekniğinin güvenilir sonuçlar verdiği belirlenmiştir.

Abstract
In this study, 26 walnut cultivars and selections were characterized and genetic relationships among them were determined by RAPD markers. DNA purity indicators were 260/280:1.87 and 260/230:2.0. DNA quantity was between 12.3 and 84.1 ng/µl (44.6 ng/µl in average). 44 Operon primers produced 301 reproducible and clear polymorphic bands. Number of bands per primer was between 2 and 17 (6.85 in average). 18 genotypes were characterized by a unique RAPD band while 8 genotypes were characterized by combination of 2-3 bands. The Dice genetic coefficient was between 0.42 and 0.94. The results showed that Kaplan-86 and Y1 were the most closely related while 65/7 and Hartley were the most distantly related genotypes. UPGMA Dendogram clustered the genotypes into three groups. It was clear that the RAPDs were reliable as long as the DNA quality was good and amplification conditions were optimized.
