Bu çalışma kapsamında ülkemizde yetiştiriciliği yapılan önemli üzüm çeşitleri ve anaçları ile bunların klonlan virüsler açısından testlenmiş ve arındırılmıştır. Asmada sertifikalı fidan üretiminde esas Arabis mosaic nepovirus, Grapevine fanleaf nepovirus, Grapevine fleck virus, Grapevine virus A, Grapevine leafroll closterovirus -1,-2,-3,-6,-7, Raspberry ringspot nepovirus, Strawbeery latent ringspot nepovirus, Tomato black ring nepovirus yönünden testlemeler yapılmış ve testlenen materyalin bir kısmı GFkV, GLRaV-1,2,3, GLFV açısından bulaşık olarak bulunmuştur. Virüslerle bulaşık olduğu belirlenen materyalden hazırlanan kalemler ve çelikler 10 litrelik plastik saksılara dikilmiş ve köklendirilip fidan elde edilmiştir. Bunu takiben bu asmalar 8 saat süreyle %60-70 oransal nem ve 30℃ sıcaklıkta ve karanlıkta, 16 saat süreyle, %60-70 oransal nem, 38℃ sıcaklıkta ve 4000-5000 lüx aydınlatmalı iklim rejiminde 40 gün süreyle termoterapi uygulamasına tabi tutulmuştur. Termoterapiyi takiben, materyalden alınan sürgün uçlarından meristem kültürü yapılmıştır. Çalışma sonucunda bazı virüslerle bulaşık olduğu belirlenen toplam 31 üzüm çeşit ve klonunun söz konusu virüslerden arındırıldığı tespit edilmiştir.
In this study, the important grape varieties, rootstocks and its clones that produced in Turkey were tested against viruses and the infected material were purged. Materials were tested against to Arabis mosaic nepovirus, Grapevine fanleaf nepovirus, Grapevine fleck virus, Grapevine virus A, Grapevine leafroll clostero virus -1,-2,-3,-6,-7, Raspberry ringspot nepovirus, Strawberry latent ringspot nepovirus, 'Tomato black ring nepovirus that are the main object of seedlings certification system. It is stated that, 31 grape varieties, rootstocks and its clones were infected with GFkV, GLRaV-1,2,3 and GLFV. After this the infected materials are subjected to purification applications. In this purpose, firstly, cuttings are planted in to the 10-liter plastic pots and they were left grow until seedlings were obtained. Then the materials were subjected to thermotherapy. Thermotherapy treatments applied to this material for 40 days with 38℃ and 4000-5000 lux lighting for 16 hours, and 30℃ and dark climate conditions. The humidity was 60-70% all the time during the study. Meristem culture followed the thermotherapy applications. Then, all of the 31-grape variety, rootstocks and clones were tested against to the viruses that mentioned above. It is determined that, all the materials were uninfected with viruses at the end of the study.
