Determination of the presence of Clavibacter michiganensis subsp. michiganensis on tomato seedlings used in Tokat province

Abstract

This study aimed to determine Clavibacter michiganensis subsp. michiganensis (Cmm) caused tomato bacterial cancer and wilt disease on tomato seedling used by farmers in Tokat province. In 2013, 2015, 2016, 2017 and 2018 years 104, 82, 94, 102 and 169 tomato seedling samples were taken from seedling providers located in Central, Turhal, Pazar, Niksar ve Erbaa in Tokat, respectively. The disease agent was detected on tomato seedling by specific polymerase chain reaction (PCR) assay after streaking on semiselective medium and extracting DNA of the bacteria growing on the medium. In 2013, 2015 and 2016, 19 out of 104, 7 out of 82 and 4 out of 94 tomato seedlings were infected with pathogen, respectively. On the other hand the disease agent was not detected in tomato seedlings samples collected in 2017 and 2018 years. According to results of this study, in seedling entered to the province, contamination ratio with Cmm decreased at important level.

Abstract

Bu çalışma,
üreticiler tarafından Tokat ilinde kullanılan, hastalık belirtisi göstermeyen
domates fidelerinde Domates bakteriyel kanser ve solgunluk hastalığı etmeni Clavibacter
michiganensis
subsp. michiganensis (Cmm)’in varlığının belirlenmesi amacıyla yapılmıştır. Tokat
Merkez, Turhal, Pazar, Niksar ve Erbaa ilçelerinden 2013 yılında 104, 2015
yılında 82, 2016 yılında 94, 2017 yılında 102 ve 2018 yılında 169 adet domates
fide örneği toplanmıştır. Toplanan örneklerden hazırlanan süspansiyonlar yar
ı
seçici besi yerine ekilmiş, gelişen bakteriler toplanarak DNA izolasyonundan
sonra etmene spesifik primerler ile polimeraz zincir reaksiyonu (PCR) analizi
yapılmıştır. Yapılan çalışma
sonucunda, 2013 yılında Tokat Merkez ve ilçelerinden toplanan 104 fide örneğinin
19’unda, 2015 yılında 82 fide örneğinin 7’sinde, 2016 yılında 94 fide örneğinin
4’ünde etmene rastlanılırken, 2017 yılında toplanan 102 fide örneğinde ve 2018
yılında toplanan 169 fide örneğinde hastalık etmenine rastlanılmamıştır. Bu
çalışma sonuçlarına göre, bölgeye giriş yapan fidelerde Cmm ile
bulaşıklık oranı önemli düzeyde azalmıştır.
