Wistar Sıçanlarında (Rattus norvegicus) Monosodyum Glutamat ile İndüklenen Uterus Tümörü Üzerinde Ganoderma mbrekobenum'un Antitümör Aktivitesi

Abstract
Mantar, Ganoderma türünün yaygın olarak antikanser ve antitümör olduğu bildirilmektedir, ancak rahim tümörleri hakkında yerli türler hakkında bir rapor yok gibi görünmektedir. Monosodyum glutamatın (MSG) yüksek konsantrasyonlarda rahim tümörlerini indüklediği gösterilmiştir. Wistar cinsi sıçanlarda bir Ganoderma türünün sulu ekstraktının MSG verilen antitümör sonuçları elde edildi. Wistar sıçanları, hayvanlarda uterus tümörünü indüklemek için monosodyum glutamat (MSG) ile beslendi ve Ganoderma mbrekobenum'un su özütü ile muamele edildi. Sıçanlara hematolojik, biyokimyasal, histopatolojik incelemeler yapıldı ve akut toksisite testi (LD50) yapıldı. Sonuçlar, tedavi edilen tüm gruplardaki sıçanlarda kilo alımının gözlendiğini göstermiştir. Böbrek ve uterus, ağırlığı artan grup 4 ve 6 dışındaki tüm gruplarda kilo almadı. MSG ile beslenen gruplarda kalp ve karaciğer ağırlığında anlamlı bir artış vardı, ancak G. mbrekobenum ile tedavi edilen grupların ağırlığı azaldı. Monosodyum glutamat ile tedavi edilen grup, hayvanlarda anemi ve toksisite belirtileri gösterdi. Sıçanların böbrek fonksiyonları ve lipid profilleri oldukça değişirken, karaciğer fonksiyonları önemli ölçüde (p< 0.05) değişti. Ayrıca, sıçanlarda östrojen seviyesi yükseldi. G. mbrekobenum ile tedavi edilen hayvanlar, sıçanlarda MSG'nin etkilerinden iyileşme belirtileri gösterdi. Akut toksisite testi, hayvanların ölümünü kaydetmedi ve G. mbrekobenum özütünün, hem MSG hem de G. mbrekobenum özütü ile beslendikten sonra sıçanların östrojen seviyelerini kontrole kıyasla önemli ölçüde düşürdüğü bulundu. Histolojik incelemede sadece MSG ile beslenen sıçanların uterus dokularında büyüme/kist varlığı görülürken, G. mbrekobenum ve monosodyum glutamat (800mg/kg/vücut ağırlığı) ile beslenen sıçanlarda histopatolojik değişiklik belirtisi görülmedi. Yukarıdaki sonuçlar, G. mbrekobenum'un Wistar sıçanlarında uterus tümörünü önleyebileceğini ve kontrol edebileceğini göstermektedir.

Abstract
Ganoderma species is widely reported to be anticancer and antitumour but there seems to be no report on indigenous species against uterine tumours. Monosodium glutamate (MSG) has been shown to induce uterine tumours in high concentrations. Antitumour effect of the water extract of a Ganoderma species on Wistar rats given MSG was conducted. Wistar rats were fed with monosodium glutamate (MSG) to induce uterine tumour in the animals and were treated with water extract of the Ganoderma mbrekobenum. Heamatological, biochemical, histopathological examinations and acute toxicity test (LD50) were done on the animals. The heart and liver in groups fed with MSG had significant increase in weight but the groups treated with G. mbrekobenum reduced in weight. The MSG-treated group showed indications of anaemia and toxicity in the animals. Renal functions and lipid profiles of the rats were fairly altered while the hepatic functions were significantly (p< 0.05) altered. Also, the amount of estrogen in the rats was elevated. Animals exposed to G. mbrekobenum showed signs of recovery from the effects of MSG in the rats. The acute toxicity test showed no fatalities among the animals and the extract of the G. mbrekobenum was notably lowered the levels of estrogen in the rats. The histological experiment revealed the presence of growths/cysts in the uterine tissues of rats fed with MSG alone, whereas the rats treated with G. mbrekobenum (800mg/kg/body weight) exhibited no histopathological alterations. These findings indicate that the G. mbrekobenum could prevent and control uterine tumour in Wistar rats.
