Evaluation of the Effects of Rheum ribes Extract on the Viability of Human Periodontal Ligament Fibroblasts: A Pilot In Vitro Study

Abstract

Objective: The aim of this in vitro study was to evaluate the effects of different concentrations of Rheum ribes extract on the cell viability of human periodontal ligament fibroblasts (hPDLF). Materials and Methods: Extracts obtained from the young shoots of the Rheum ribes plant were prepared at three different concentrations (5, 25, and 50 μg/mL). Each concentration was evaluated in five replicates for human periodontal ligament fibroblasts (hPDLF). Cells were treated with the extracts for 24 hours, after which cell viability was measured using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Data were analyzed using JASP statistical software, and the level of statistical significance was set as p < 0.05. Results: No statistically significant effect on cell viability was found with the 5 and 25 μg/mL concentrations of the Rheum ribes extract. In contrast, the 50 μg/mL concentration demonstrated significant cytotoxicity in hPDLF cells. Conclusion: These findings indicate that the 5 and 25 μg/mL concentrations of Rheum ribes extract did not have a significant effect on cell viability, whereas the 50 μg/mL dose demonstrated significant cytotoxicity. Overall, the study underlines the importance of dose optimization and provides a preliminary basis for future research on the biological potential of this plant extract.

Rheum ribes Ekstresinin İnsan Periodontal Ligament Fibroblastlarının Canlılığı Üzerindeki Etkilerinin Değerlendirilmesi: Bir Pilot İn Vitro Çalışma

Abstract

Amaç: Bu in vitro çalışmanın amacı, Rheum ribes bitkisinden elde edilen farklı dozlardaki ekstrelerin insan periodontal ligament fibroblastlarının hücre canlılığı üzerindeki etkilerini değerlendirmektir. Gereç ve Yöntemler: Rheum ribes bitkisinin genç sürgünlerinden elde edilen ekstreler 5, 25 ve 50 μg/mL olmak üzere üç farklı konsantrasyonda hazırlandı. İnsan periodontal ligament fibroblastları (iPDLF) için her bir konsantrasyon beş tekrar ile değerlendirildi. Ekstreler hücrelere 24 saat uygulandı ve ardından hücre canlılığı 3-(4,5- dimetiltiazol-2-il)-2,5-difeniltetrazolyum bromür (MTT) analizi ile ölçüldü. Veriler JASP istatistik yazılımı kullanılarak analiz edildi ve istatistiksel anlamlılık düzeyi p<0.05 olarak kabul edildi. Bulgular: Rheum ribes bitkisinden elde edilen 5 ve 25 μg/mL konsantrasyonlarındaki ekstrelerin hücre canlılığı üzerinde istatistiksel olarak anlamlı bir etkisi saptanmadı. Buna karşılık, 50 μg/mL konsantrasyondaki ekstrenin hücreler üzerinde sitotoksik etki gösterdiği bulundu. Sonuç: Bu çalışmada edinilen bulgulara göre, Rheum ribes ekstresinin 5 ve 25 μg/mL konsantrasyonlarında hücre canlılığı üzerinde anlamlı bir etki göstermediğini, buna karşılık 50 μg/mL dozunda belirgin sitotoksisite oluşturduğunu ortaya koymaktadır. Genel olarak çalışma, doz optimizasyonunun önemini vurgulamakta ve bu bitki ekstresinin biyolojik potansiyeline yönelik gelecekte yapılacak araştırmalar için öncü nitelikte bir temel sunmaktadır.
