KUZULARDA DENEYSEL OLARAK OLUŞTURULAN SEREBROKORTİKAL NEKROZDA KLİNİK, LABORATUVAR VE PATOLOJİK BULGULAR

Abstract

Bu çalışma, kuzularda amprolium ile deneysel olarak serebrokortikal nekroz (CCN) oluşturmak ve buna ilişkin
gelişen santral sinir sistemindeki patolojik değişiklikleri belirlemek amacıyla yapıldı. Materyal olarak 7-9 aylık, canlı ağırlıklan
11·15 kg arasında olan 6 kuzu kullanıldı. Kuzular 3'erli 2 gruba ayrıldı . Bütün kuzulara 600 mg/kg dozunda amprolium
soıusyonu sonda ile içirildi. Kuzularda CCN 29·63 günler arasında oluştu . Grup l'deki kuzulara herhangi bir tedavi
uygulanmazken, grup ll'deki kuzulara, serebrokortikal nekroz belirtileri gözlendikten sonra tiamin (500 mg/gün) 4
gün süre ile uygulandı. Plazma tiamin seviyesi 4.42 J.Jg/ml'den 0,62 J.Jg/ml'ye (p<0.01) düştüğü ve tiamin tedavisi (ll.
grup kuzularda) sonrası 3,2 J.Jg/ml'ye yükseldiği belirlendi. Serum CPK, ALT, Glikoz ve AST konsantrasyonlannda
önemli (p<0.05) artışlar kaydedildi. Grup l'deki kuzular CCN belirtileri gözlendikten sonra, grup ll'deki kuzular ise tiamin
tedavisinin bitiminden sonra kesilerek otopsileri yapıldı. makroskopik olarak, grup l'deki kuzuların hepsinin meninksleri
hiperemik, beyin dokusu şişkin ve ödemli idi. Mikroskopik olarak ise parivasküler ve parinöral ödem, işemik
neural değişiklikler, serabral-kortekste fokal nekrotik odaklar, kollikuluslarda kanamalar tespit edildi. Sonuç olarak,
amproliumun uzun süreli ve yüksek dozda kullanıldığında kuzularda CCN'a yol açtığı ve tiamin yetmezliği saptanan kuzuıara
tiamin uygulandığında tedavi uygulandığında tedavi edici sonuç alınabileceği kanısına varıldı.

Clinical, Laboratory and Pathological Findings in Cerebrocortical Necrosis (CCN) Induced Experimentally in the Lambs

Abstract

This study was undertakan to determine pathological changes of cerebrocortical necrosis (CCN) experi·
mentally induced by given amprolium in lambs. Six healthy lambs. aging between 7 and 9 months and weighting between
11 and 15 kg were used as a material. Six lambs were divided into two groups to be three animals in each
group (group 1,11). Amprolium ( 600 mg/kg/ day) was given per oral to the both groups of lambs by stomach tube during
experiment. The symptoms of CCN in lambs were observed between 29 and 63 days of the experiment. While the
lambs in the group ll were treated with thiamine ( 500 mg /day) after elinical symptoms of CCN were appeared, the
ıambs in the group ı were not applied any treatment. Plasma tiamin concentration decreased dramatically from 4.42
J.Jgfml to 0.62 J.Jglml (P< 0.01) and altering treatment, inclined to 3.2 J.Jg/ml in the lambs of group ll. Serum CPK, ALT,
AST ve glucose concentration increased dramatically (p<0.01 ). The lambs in the group ı were necropsied when the
all symptoms of CCN were observed. The lambs in the group ll were necropsied when the elinical symptoms of CCN
were disappered after treatment. At the necropsy, meningial hiperemia, swollen and oedematus brain tissues were
observed in all the lambs of group ı. The slight oedema in the brain was detected in the lambs group ll. Histologically,
ichemic neuronal changes , perivasculer and perineuronal oedema. focal necrosis in carebellar cortex and colliculus
were determined in all the lambs of group 1. The results of this research work showed that using long term and high
dose of amprolium in lamb caused cerebrocortical necrosis and deficies thiamin of lambs with thiamin treatment would
be of benefit in the care of CCN.
