The Investigation of Effects of Vitamın C and Benfothiamine on Alterations in The Experimental Diabetic Rat Kidney Tissues

Abstract

Objective: The protective effects of Benfothiamine and Vitamin C against apopitotic changes in streptozotosin (STZ) induced experimental diabetic rats were investigated. Materials and Methods: Twenty eight rats were included in the study. The rats were divided into 4 groups as 7 rats in each group. Control group, had no intervention throughout the study. To induce diabetes mellitus (DM) other 3 groups were administered 0.1 M STZ in sodium citrate buffer intraperitoneally. DM group, received no other medication until the end of the study. DM + Benfothiamine group received 70 mg/kg/day Benfothiamine by oral route for a period of 6 weeks. DM+Vitamin C group received 900 mg/kg/day Vitamin C by oral route for a period of 6 weeks. In the end of the experiment, rats were decapitated, renal tissues were obtained and stained with hematoxylen-eosin and Tunel paints. Results: DM+Benfothiamine group showed mesengial matrix hypertrophy in glomerules, although the increasement was less than the DM group. DM+Vitamin C group also showed increased mesangial matrix and glomerular hyperthrophy. DM+Vitamin C group showed less significant tubuler dilatation, dissociation of tubuler epithelium, and glucogenic vacuolization. This protective effect was not as much as the DM+Benfothiamine group. When compared to control group, DM group had significantly increased Tunel positivity. DM+Benfothiamine and DM+Vitamin C group showed decreased Tunel positivity in comparasion to the DM group. Conclusion: Benfothiamine and Vitamin C were found to have protective effects on the renal dysfunction induced by DM.

Benfotiamin ve C Vitamininin Deneysel Diyabetik Sıçan Böbrek Dokusundaki Değişiklikler Üzerine Etkilerinin Araştırılması

Abstract

Amaç: Bu çalışmada, Streptozotosin (STZ) ile oluşturulan deneysel diyabet modelinde Benfotiamin ve Vitamin C'nin sıçan böbrek dokusundaki apoptotik değişiklikler üzerine koruyucu etkileri incelenmiştir. Gereç ve Yöntem: Çalışmada 28 adet 6 haftalık Wistar albino cinsi erkek sıçanlar kullanıldı. Deney hayvanları her grupta 7 hayvan olacak şekilde 4 gruba ayrıldı. Kontrol grubuna herhangi bir işlem yapılmadı. Diğer 3 gruba 50 mg/kg olacak şekilde tek doz STZ 0,1 M sodyum sitrat tamponunda çözdürülerek intraperitoneal olarak verildi. Diyabetik grup; sadece kan şekeri takibi yapıldı. DM + Benfotiamin grubuna; benfotiamin 70 mg/kg/gün ve DM + Vitamin C grubuna Vitamin C (900 mg/kg/gün) 6 hafta süreyle oral olarak verildi. Deney sonrası sıçanlar dekapite edilerek böbrek dokuları çıkarıldı. Böbrek dokularına Hematoksilen- Eozin ve TUNEL boyama yapıldı. Bulgular: Benfotiamin grubuna ait böbrek dokularında, diyabetik grubdan daha az miktarda glomerüllerde mezengial matriks artışı ve hipertrofi gözlendi. Vitamin C grubuna ait böbrek dokularında glomerüllerde diyabetik gruptakine benzer bir şekilde belirgin mezengial matriks artışı ve glomerüler hipertrofi gözlendi. Vitamin C grubuna ait böbrek dokusunda tübüllerde ise diyabetik grupla karşılaştırıldığında daha az belirgin oranda tübüler dilatasyon, tübül epitellerinde ayrılmalar ve glukojenik vakuolizasyon izlendi. Ancak bu düzelme Benfotiamin grubu ile karşılaştırıldığında daha az belirgindi. Kontrol grubu ile kıyaslandığında diyabetik grupta belirgin olarak artmış TUNEL pozitifliği tespit edildi. Benfotiamin ve Vitamin C gruplarında ise TUNEL pozitifliğinin diyabetik gruba göre anlamlı olarak azalmış olduğu gözlendi. Sonuç: DM'nin yol açtığı hücresel hasara bağlı olarak gelişen böbrek fonksiyon bozukluklarına karşı Benfotiamin ve C Vitamininin koruyucu etkilerinin olduğu gözlendi.
