Detection of Viruses in Aegean Region Grapevines

Abstract

This study was conducted to determine virus diseases on grapes in İzmir
and Manisa provinces in 2007-2010 using serological and molecular methods. In
addition to this,detection and identification of grapevine viruses on samples
from Çanakkale and Denizli in 2009-2010 were performed.

To identify of viruses in grapevine production areas in the four provinces,
the samples were collected from plants showing virus symptoms. Sampling was
done 2 times a year in the spring and in the autumn. In the spring, shoot and
leaf samples were taken from annual shoots of plants with virus like symptoms
(yellow mosaic,vein clearing, ringspot, vein banding, fasciations, zigzag
growth, shortening of internodes, abnormal branching). In the autumn, reddish
and rolling downwards leaf samples from annual shoots were collected. Totally
746 samples were taken from plants with virus like symptoms in surveyed area in
2007-2010.





Identification of viruses was performed using serological and molecular
methods. DAS-ELISA test results showed that the plants were infected with GFLV,
GFkV, ArMV, GLRaV 1, 2, 3 and 4-9 viruses. PCR analysis confirmed DAS-ELISA
results. In PCR tests, bands for GFLV, GFkV, ArMV, GLRaV 1, 2, 3 and 4 were
obtained.
