Nohutta Kök Çürüklüğü ve Solgunluğa Neden Olan Fusarium spp. İzolatlarının In Vitro Koşullarda Virülenslik Derecelerinin Belirlenmesi

Abstract

Bu çalışma, Eskişehir, Uşak, Kütahya ve Denizli illerinde nohutta kök çürüklüğü ve solgunluğa neden olan Fusarium türlerinin elde edilmesi ve virülenslik derecelerinin belirlenmesi amacıyla yapılmıştır. Yapılan surveyler sonucunda, Eskişehir’de 12 tarladan 225 bitki, Uşak’ta 16 tarladan 243 bitki, Kütahya’da 16 tarladan 308 bitki ve Denizli’de 12 tarladan 116 bitki olmak üzere toplamda 56 farklı tarladan 892 adet kök çürüklüğü ve solgunluk belirtisi gösteren bitki toplanmıştır. Bu bitkilerden yapılan izolasyonlar sonucunda, Kütahya’dan toplanan bitkilerden 178, Eskişehir’den 147, Denizli’den 167 ve Uşak’tan 222 izolat olmak üzere toplam 714 adet Fusarium spp. izolatı elde edilmiştir. 2016 yılında in vitro koşullarda bu izolatların patojenisite testleri yapılmış ve % hastalık şiddetine göre virülenslik dereceleri belirlenmiştir. Toplam 714 Fusarium spp. izolatı arasından 32 tanesinin %0-20, 113 tanesinin %20-40, 134 tanesinin %40-60, 271 tanesinin %60-80 ve 164 tanesinin ise %80-100 arası hastalık şiddeti değerine sahip olduğu saptanmıştır. Bu değerlere göre virülenslikleri yüksek izolat oranı % 60,9 olarak belirlenmiştir.

Determination of Virulence Grades of Fusarium spp. Isolates Causes Wilt and Root Rot in Chickpea In Vitro Conditions

Abstract

This study was carried out in order to determine the virulence grades of Fusarium species causing root rot and wilt on chickpea in Eskişehir, Uşak, Kütahya and Denizli provinces by pathogenicity tests. In this context, firstly, survey studies were carried out during the 2015 chickpea production season. As a result of the surveys, a total of 892 plants showing disease symptoms were collected from 56 different fields, including 225 plants from 12 fields in Eskisehir, 243 plants from 16 fields in Uşak, 308 plants from 16 fields in Kütahya and 116 plants from Denizli. As a result of isolations made from these plants, a total of 714 Fusarium isolates were obtained, 178 of which were collected from Kütahya, 147 from Eskişehir, 167 from Denizli and 222 isolates from Uşak provinces. In 2016, the isolates were tested for pathogenicity in petri dishes in vitro conditions, and their virulence grades were determined based on the percentage of disease severity. The results showed that, 32 isolates caused disease severity rates between 0-20%, 113 isolates between 20-40%, 134 isolates between 40-60%, 271 isolates between 60-80% and 164 isolates between 80-100%. According to these results, high virulence isolate rate was determined as 60,9% of all isolates.
