SIÇAN OLFAKTOR EPİTELİNDE KOLŞİSİN İNDÜKLÜ APOPİTOZİSİN P-53 İLE VE YENİLENMESİNİN PCNA İLE GÖSTERİLMESİ

Abstract




Olfaktor epitelde, sıçan
proteinleri için özel poliklonal antikorlar kullanılarak immünohistokimya
gerçekleştirildi. Apopitozis için p-53 ve hücre proliferasyonu için PCNA (proliferating cell nuclear antigen)
ile sıçan proteinleri incelendi. Ayrıca olfaktor epitel hematoksilen-eozinle
boyanarak tüm yapı gözden geçirildi.




 




Yüksek tek doz (i.p) kolşisin
(6mg/kg) sıçanlara uygulandı. Uygulamadan 1 gün sonra, PCNA immün boyaması,
olfaktor epitelin bazal hücrelerinde negatif 
bulundu oysa epiteldeki apopitotik hücreler p 53 için pozitifdi.
Apopitozis immunoreaktivitesi, sitotoksik ajan kolşisin uygulandıktan sonra, en
erken 1. günde görüldü. 1. ve 2.Günde, olfaktor epitelin bazı alanlarının bazal
bölge hücrelerinin tümü, birkaç yuvarlak bazal hücreler ve Bowman bezlerinin
kanal hücreleri hariç ölmüştü. 3.günde, hücre ölümüne cevap olarak, olfaktor
epitelde proliferasyon prekürsör hücrelerde (yuvarlak bazal hücreler) ve
ardından olfaktor hücrelerde rejenerasyon meydana geldi. 5-7. Günlerde,
rejenerasyon doruğa ulaşmıştı, hücreler kontrolden daha fazla arttığı PCNA
immün boyamasıyla belirlendi. 10-15. Günlerde, rejenerasyon meydana geldi,
epitel normal görünüyordu ancak bazı bölgelerde, nadiren apopitotik hücreler
gözlendi. Uygulamanın 15. gününde tam olarak rejenerasyon oluşmuştu. Bu
çalışma, olfaktor epiteldeki bazal hücrelerin, epiteli yenileyebileceğini ve
tam bir rejenerasyon oluşumunun  zamana
bağlı olacağını göstermektedir.
