Bazı Kırmızıbiber Hatlarının Patates Y Virüs (Potato Virus Y, Pvy) Patotiplerine Reaksiyonları

Abstract

Bu çalışmada, baharat yapımına uygun aday 43 kırmızıbiber hattının Patates Y virüsü (PVY)'nin (0), (0,1), ve (0,1,2) patotiplerine reaksiyonları araştırılmıştır. PVY’nin, LYE84 (0), CAA16 (0,1) ve SON41P (0,1,2) izolatları Nicotiana tabacum L. " Samsun " tütün çeşidi üzerinde çoğaltılmış ve virüsün varlığı PVY poliklonal antiserum kullanılarak DAS-ELISA testi ile doğrulanmıştır. Daha sonra patotiplerin pvr lokusuyla ilişkileri Yolo Wonder, Yolo Y, Florida VR2 ve W4 biber genotiplerinde biyolojik ve serolojik olarak kontrol edilmiştir. Denemeye alınan kırmızıbiber hatlarına, PVY patotipleri mekanik olarak inokule edilmiştir. Biber hatlarında PVY patotiplerinin çoğalması ve yayılması, pozitif kontrollerde ilk simptomları takiben DAS-ELISA testiyle üç hafta süreyle izlenmiştir. PVY (0) patotipi, bitki yapraklarında mozaik, PVY (0,1) patotipi, yaprak yüzeyinde deformasyon, yapraklarda içe kıvrılma, bitki gövdesinde şekil bozukluğu ve meyvede şeritler halinde renk açılması simptomları oluşturmuştur. PVY (0,1,2) patotipi, yaprak damarı çevresinde nekrozlar oluşturmuş ve yapraklar dökülmüştür. Üç kırmızıbiber hattında PVY patotiplerinden kaynaklı simptom meydana gelmemiştir. Serolojik testlerden alınan sonuçlara göre, bu üç hattın, PVY’nin üç patotipine dayanıklı aday hatlar olduğunu söylemek mümkündür.
