Development of Eggplant (Solanum melongena L.) Breeding Lines Resistant to Root-Knot Nematode and Fusarium Wilt Using Molecular Markers

Abstract

Root-knot nematodes (Meloidogyne spp.) and Fusarium wilt caused by Fusarium oxysporum f.sp. melongenae are among the most destructive soil-borne pathogens limiting eggplant (Solanum melongena L.) production worldwide. The development of cultivars with durable disease resistance has therefore become a major objective of modern eggplant breeding. This study aimed to identify resistance alleles associated with these two economically important pathogens using molecular markers in 157 advanced F₃ breeding lines developed within an eggplant breeding program. Resistance to Fusarium wilt was evaluated using the SCAR426 marker, whereas resistance to root-knot nematodes was assessed with the Rex CAPS marker linked to the Mi-1 resistance locus. Among the 148 breeding lines successfully analyzed with the SCAR426 marker, 24 (16.22%) were identified as homozygous resistant (RR), 7 (4.73%) as heterozygous resistant (Rr), and 117 (79.05%) as homozygous susceptible (rr). In the Rex CAPS marker analysis, six breeding lines produced no interpretable results; however, all 151 successfully evaluated lines carried the homozygous resistant (RR) genotype for root-knot nematode resistance. These findings indicate that resistance to root-knot nematodes has been effectively fixed within the breeding population, whereas resistance to Fusarium wilt remains limited and requires further selection. Importantly, breeding lines carrying resistance alleles to both pathogens were identified, providing valuable parental materials for gene pyramiding and marker-assisted selection. The identified resistant lines represent important genetic resources for the development of eggplant cultivars with multiple disease resistance and will be further validated through phenotypic screening to support sustainable eggplant production and environmentally responsible breeding strategies.
