Relationship between adiponectin and adiponectin receptor 1 gene polymorphisms with colorectal cancer

Abstract

Objectives: Adiponectin is a protein synthesized from adipose tissue and it is also an endogeneous insulin sensitizer. Adiponectin seems to act in preneoplastic colonic lesions to regulate cell growth by activating, altering, or interacting with several pathways including the leptin and the NFkB pathways. This study aims to investigate the relationship of T/G rs2241766ADIPOQ exon 2 (+45,codon 15) and A/G rs2275738ADIPOR1 intron 1 (-106) polymorphisms with CRC risk. Methods: The studying group consist of total 266 person which 129 of them are colorectal cancer cases who were diagnosed at the Mersin University Medical Faculty Hospital and 137 controls participating. Genotypes were determined using PCR/RFLP methods. Results: It was found that there is no relationship between T/G rs2241766ADIPOQ exon 2 (+45, codon 15) and A/G rs2275738ADIPOR1 intron 1 (-106) polymorphisms and the risk of developing colorectal cancer. It was determined an increasing risk of colorectal cancer with male subjects and old age groups. Conclusion: In conclusion, it was found that rs2241766 and rs2275738 polymorphisms may not be related to the progression of colorectal cancer. J Clin Exp Invest 2014; 5 (4): 572-576

Adiponektin ve adiponektin reseptör 1 gen polimorfizmleri ile kolorektal kanser arasındaki ilişki

Abstract

Amaç: Adiponektin adipöz doku tarafından sentezlenen bir proteindir ve endojenik insülin duyarlayıcısıdır. Adiponektinin preneoplastik kolonik lezyonlarda hücre büyümesini aktive ederek, değiştirerek veya leptin ve Nükleer Faktör Kabba B (NFkB) yollarını da içeren birkaç farklı yolla kolorektal kanser (KRK) gelişimini etkilediği düşünülmektedir. Bu çalışmada T/G rs2241766ADIPOQ exon 2 (+45,codon 15) ve A/G rs2275738ADIPOR1 intron 1 (-106) polimorfizmleri ile kolorektal kansere yakalanma riski arasında ilişki olup olmadığının araştırılması amaçlanmıştır. Yöntemler: Çalışma grubu, Mersin Üniversitesi Tıp Fakültesi Hastanesi\'nde KRK tanısı almış 129 hasta ve 137 kontrol grubu olmak üzere 266 bireyden oluşturuldu. Her bir birey için PCR/RFLP yöntemleri kullanılarak genotipler belirlendi. Bulgular: Adiponektin (ADIPOQ) geninin +45,codon 15 pozisyonundaki T/G değişimi ve Adiponektin Reseptör 1 (ADIPOR1) geninin intron 1 (-106) pozisyonundaki A/G değişimini içeren polimorfizmlerin kolorektal kansere yakalanma riskini etkilemediği saptanmıştır. KRK\'ya yakalanma riskinin erkek bireylerde ve ileri yaş gruplarında arttığı tespit edilmiştir. Sonuç: T/G rs2241766ADIPOQ ekzon 2 (+45,codon 15) ve A/G rs2275738ADIPOR1 intron 1 [-106] polimorfizmleri ile kolorektal kanser arasında ilişki olmadığı belirlenmiştir.
