Abstract

Bu çalışmada trisiklik antidepresan ilaçlardan amitriptilin ve
nortriptilinin idrar ve plazmadan pH = 11 de n-heksan ile ekstraksiyonu yapılmış ve İ.T.K. tarama testi olarak kullanılmıştır. Silikagel-G adsorban tabakasında (etil asetat: n-heptan:metil alkol: amonyak 60:30:7.5:7.5:) developman sistemi ile en iyi ayrılma sağlanmış ve Dragendorff reaktifi ile duyarlık alt sınırı 1 g olarak saptanmıştır. Destekleyici analiz ve miktar tayini ise GLK ile yapılmıştır. PackardBecker 419 gaz kromatografında % 3 OV-17 (Chromosorb w-HP 100 -120 mesh üzerinde) kolonu ve FID detektörü kullanılmıştır. Duyarlık amitriptilin ile 0.5 g/ml vücut sıvısı, nortriptilin ile 1 g/ ml vücut sıvısı olarak saptanmıştır. verim ise amitriptilin ve nortriptilin ile sıra ile % 62 1.3 ve % 74.3 1.5 elde edilmiştir.

Abstract

Thin-Layer Chromatagraphy was used as a serening test in this
research for urine and plazma extracts containing amitripthyline and
nortriptyline at concentrations 5,25, 50 and 100 g / 5 ml biologic
fluid. Extraction was made with n- heptane at pH 11. The developing
system (ethyl ecetate: n-heptane: methyl alcohol:ammonia: 60:30:
7-3:7-5) we used on Silicagel-G plates gave the test separation and 1
g drug can be detected using Dragendorff reagent.
Gas-Liquid Chromatography (GLC) was used for confirmation
and for the quantitative determination of the drugs in the extract.
Analyses were performed on Packard-Becker 419 Gas Chromatograph
equipped with FID and glass column 2 m X 4 mm (i.d) packed
with 3% OV-17 on Chromosorb W-HP, 100-120 mesh and protriptyline
was used as an internal standart. The lower limit of sensitivity
with GLC was 0.5 g/ml biologic fluid for amitriptyline and 1
g/ml biologic fluid for nortriptyline.
The receveries were 62 1.3 % for amitriptyline and 74.3 1.5
% for nortriptyline.
