Abstract

Bu çalışmada, Manisa Spil dağından toplanmış olan Centaurea cyanus’un kurutularak toz edilmiş toprak üstü kısımlarından, n-hekzan, kloroform ve metanol ekstreleri hazırlanmıştır. Çeşitli kromatografik yöntemler kullanılarak n-butanol ekstresi üzerinde fraksiyonlama ve izolasyon işlemleri gerçekleştirilmiştir. İzole edilen saf maddelerin yapı tayinleri spektroskobik yöntemler (1H ve 13C NMR) kullanılarak gerçekleştirilmiştir. Bazı fraksiyonlarda ticari saf standart maddeler kullanılarak İTK ile analiz yapılmıştır. C. cyanus’tan elde edilen n-BuOH ve MeOH ekstrelerinin içerikleri UPSK ile flavonoit bileşikleri ve klorojenik asit içeriği açısından incelenmiştirr. Sonuç olarak, C. cyanus’tan izoramnetin-7-O- -Dglukopiranozit ve apiin bileşikleri izole edilmiştir. Siringin, klorojenik asit ve apigenin varlığı ise İTK ile tespit edilmiştir. Ayrıca, ekstrelerde klorojenik asit, homoorientin, luteolin-3-O-glikozit ve luteolin-4-Oglikozit bulunduğu UPSK metodu ile belirlenmiştir. Bitkide siringin, homoorientin, luteolin-3-O-glikozit ve luteolin-4-O-glikozit varlığı bu çalışma ile ilk kez ortaya konmaktadır

SECONDARY METABOLITES OF CENTAUREA CYANUS L

Abstract

In this study, n-hexane, CHCl3 and MeOH extracts from dried and powdered aerial parts of C. cyanus collected from Manisa, Mountain Spil were prepared. Fractionation and isolation procedures were performed on the n-BuOH extract by using several chromatographical methods. Structure elucidation of the pure compounds were performed by using spectroscopic methods (1H and 13C NMR). Identification studies were performed with TLC by using commercial pure standarts on some fractions. Contents of the n-BuOH and MeOH extracts prepared from C. cyanus were determined by comparing flavonoids and chlorogenic acid standarts on UPLC instrument. As a result, isorhamnetin-7-O- -D-glucopyranoside and apiin were isolated from C. cyanus. Syringin, chlorogenic acid and apigenin were identified by using a TLC method. Furthermore, chlorogenic acid, homoorientin, luteolin-3-O-glycoside and luteolin-4-O- glycoside were determined in the extracts by using an UPLC method. Syringin, homoorientin, luteolin-3-O-glycoside and luteolin-4-O-glycoside were reported for the first time in C. cyanus by our work
