THE INVESTIGATION OF SOME ENZYME LEVELS IN THE KIDNEY, SPLEEN, HEART, LIVER AND BRAIN OF DAMİNOZİDE ADMİNİSTERED RATS

Abstract

In this study, it was aimed to examine possible changes in enzyme activities and total protein levels in tissues of rats given daminozit at the dose of 150 mg/ kg/ day for 15. days introperitonly. Rats were killed by cervical dislocation on the end of 15 day . total protein (TP), alanin amino transferase (ALT), laktate dehidrogenase (LDH), alkaline fosfatase (ALP), gamma glutamil transpeptidaz (GGT), aspartat amino transaminaz (AST) enzyme actıvıtıes were measured In prepared homogenates of spleen, kidney, heart, liver and brain tissues by Ciba Corning Express Plus autoanalyzer. GST enzyme was studied by spectrofotometric assay. The data of experimental group was compared with those of control rats. There were no statisticallyt differences between experimental group and control group in terms of enzymes in liver and kidney (p>0,05) . There were significant differences in experimental group compared to control rats in spleen TP (u=32;p<0,05) and GGT (u=36 ;p<0,05) in heart GST (u=36;p<0,05) and brain GST (u=36;p<0,05) and GGT (u=32;p<0,05) enzymes (p<0,05), but the other enzyme activities did not show significant differences between two group (p>0,05)

DAMİNOZİT UYGULANAN SIÇANLARIN BÖBREK, DALAK, KALP, KARACİĞER VE BEYİNLERİNDE BAZI ENZİM DÜZEYLERİNİN İNCELENMESİ

Abstract

Daminozit'in total protein ve dokulardaki enzim parametreleri üzerindeki olası değişimlerini izlemeyi amaçladığımız çalışmamızda, sıçanlara 15 gün süreyle 150 mg/kg/gün dozunda daminozit intraperitonal injeksiyon ile uygulandı. Denekler 15. gün bitiminde servikal dislokasyonla öldürüldü. Böbrek, dalak , beyin, karaciğer ve kalplerinden hazırlanan homojenatlarda total protein (TP) ile alanin amino transferaz (ALT), laktat dehidrogenaz (LDH), gama glutamil transpeptidaz (GGT), alkalen fosfataz (ALP), aspartat amino transaminaz (AST) ve glutatyon -s- transferaz (GST) enzim aktiviteleri ölçüldü.. GST aktivitesi spektrofotometrik yöntemle çalışıldı diğer tüm enzim aktiviteleri ise Ciba Corning Express Plus otoanalizörde ölçüldü. Elde edilen sonuçlar, kontrol grubu sıçanların enzim aktivite sonuçları ile karşılaştırıldı. Yapılan istatistiksel analizde deneklere ait enzim aktivite ölçümleri kontrol grupları ile karşılaştırıldığında Böbrek ve karaciğerde tüm enzim aktivite değerlerinde gruplar arası farklılık anlamsız olarak bulunurken (p>0,05) . dalakta T.P (u=32;p<0,05) ve GGT (36;p<0,05) değeri hariç tüm gruplarda fark anlamsız bulundu (p>0,05), kalpte GST aktivitesi (u= 36; p<0,05), beyinde ise GST (U=36;p0,05)
