Abstract
Optimizasyon of Topical Preparations of Retinoic Acid and İn Vitro-İn Vivo Evaluation In this study, the solubility of retinoic acid, which is practically insoluble in water and sensitive to air and sunlight has been studied to increase by cyclodextrins to prepare a new topical preparations for acne treatment. For this purpose 4 different types cyclodextrins as -cyclodextrin, hydroxypropyl- -CD, randomyl methylated -cyclodextrin and -cyclodextrin were used and the inclusion complexes of retinoic acid with these cyclodextrin's were prepared. Then the solubility of the complexes were determined. The formulation of complexes were defined by the IR spectrums, DSC thermograms and X-Ray powder diffractometry analyses. According to the solubility, IR, DSC and X-Ray analyses results the inclusion complex which has prepared using -cyclodextrin by modified autoclave method in 1:1 molar ratio was choosen the most suitable compound. After that the formulation studies has been carried out with this choosen complex. The studies has been carried out on 2 types of commercial preparations which are in gel and cream form and 3 different types of hydrogel and 3 different types of moisturizing cream base formulations which is prepared by us. The release of retinoic acid from the formulations was also investigated by using modified Franz cell with cellulose acetate membrane. The hydrogel and moisturizing base prepartions has been choosen according to in vitro release profiles. The clinical studies of these two preparations have been realized on volunteer acne vulgaris patients in Ankara University, Faculty of Medicine, Department of Dermatology. It was found that, the F5B coded formulation which is contain retinoic acid- -cyclodextrin complex in moisturizing base was better and more effective than the commercial products. It was not also seen any side effect with this formulation
