Investigating the healing potential of yashtimadhu (licorice) using dental pulp stem cell

Abstract

Herbal medicine, a centuries-old practice, has gained increasing attention in the modern healthcare
landscape. Yashtimadhu is an ancient herbal medicine which has anti-inflammatory, anti-microbial and wound healing
properties. It contains phytochemical component such as glabridin, glycyrrhizin, licochalcone which are responsible for
wound healing. Our objective is to study the role of Yashtimadhu aqueous extracts (YAC) on dental pulp stem cells
(DPSCs) with respect to its wound healing action. DPSCs were isolated and characterized for surface marker CD34,
CD45, CD73, CD90, CD105 and HLA-DR. The cytotoxicity of Yashtimadhu extract was studied using MTT assay at
various concentrations like 2, 5, 10, 15, 20, 25, 50 μg/ml. Later, scratch assay was performed. Cells were treated with
Mitomycin C to inhibit cell division and further treated with aqueous extracts of Yashtimadhu to evaluate its effect on cell
migration. Scratch was observed at 6 and 24 hours and images were taken under inverted microscope. Further the image
was analyzed using image J software. Cell cycle analysis was performed using flow cytometer. Using Yashtimadhu extract
on DPSC cell lines demonstrated no cytotoxic effects and increased cell migration, aiding in wound repair. Additionally,
combining Yashtimadhu with Mitomycin C enhanced wound healing and cell migration, suggesting its potential for novel
tissue repair therapies.
