Mangiferin Attenuates Nicotine-Induced Testicular Oxidative Stress, Endocrine Disruption, and Spermatogenic Impairment in Adult Male Wistar Rats

Abstract

The consumption of nicotine (NICO) through smoking is a serious public health issue. Nicotine is associated with infertility and the altered spermatogenesis and metabolism of male reproductive hormones. Mangiferin (MANG) is a naturally occurring C-glucosyl xanthonoid compound found in Mangifera indica and has been characterized as a well-known antioxidant and anti-inflammatory compound. This study was initiated to investigate the protective role of Mangiferin in nicotine-induced testicular toxicity. The groups consisted of Control, NICO (1.5 mg/kg, i.p), NICO + MANG co-treatment (1.5 mg/kg i.p. + 25 mg/kg p.o.), NICO+MANG post-treatment (1.5 mg/kg i.p. for 32 days followed by 25 mg/kg p.o. for 32 days), and MANG (25 mg/kg p.o.). The reproductive hormones (FSH, LH, testosterone, prolactin, and estradiol) were assayed; antioxidant enzyme activities (SOD, CAT, GSH, and GSSG), lipid peroxidation (MDA), and steroidogenic enzyme (17ß-HSD, G6PDH, and hexokinase); and testicular histoarchitecture were also analyzed. Histological sections of the testes of the nicotine-induced rats revealed desquamated cells, atrophic germinal epithelium, thickened basement membrane, congested blood vessels, depleted lumina, and reduced tubular diameter, as well as increased luminal diameter compared to the normal control. The reproductive hormones were also significantly reduced compared to the control groups. Observed deleterious effects in NICO rats were generally mitigated in the post-treatment with MANG group rats. These findings indicate that MANG ameliorates the severity of toxicity induced by nicotine in rat testes.
