In order to evaluate the effect of dexamethasone treatment upon the synthesis of DNA in human central nervous system cell cultures, nine different type cell cultures were used. Twin high cell density cultures for each cell type were used and one of these was treated with 10 ugr/ml dexamethasone for one week.
These cells were investigated with FacS II (Beeton Dickinson) after treatment- with chromomycin A (Sigma lab).
At the end of the research it was seen that, there were more cells in synthesis and myosis period in astrocytomas while there was partly blocage at G1 phase, which is related to the dexamethasone treatment, in metastatic and normal cells.
insan SSS hücre kültürlerinde deksametazon tedavisinin DNA sentezi (S) üzerine etkisini saptamak amacıyla 9 değişik tip hücre kullanıldı.
Yüksek dansiteye ulaşmış kültürlerden her hücre tipi için bir kontrol ve bir de 1 haftalık 10 ugr/ml deksametazon tedavisi alan olmak üzere ikişer örnek hazırlandı. Bu hücreler chromomycin A (Sigma lab) ile muameleden sonra FacS II ile (Becton Dickinson) incelendi.
Normal hücrelerle metastik tümör hücrelerinde deksametazonla tedaviye bağlı Gı de kısmi blok tesbit edilirken astrositik tümör hücrelerinde sentez ve mitoz devresine ulaşmış hücre sayısı normal hücrelere oranla daha fazla bulundu.
