Tedavide sıklıkla kullanılan bazı elekrolitlerin insan eritrosit glukoz 6-fosfat dehidrogenaz enzimi (G6PD) üzerine olan etkileri in vitro koşullarda araştırıldı. GPD enzimi afinite kromatografisi ile saflaştırıldı. Enzim aktivitesinin tayini Beutler metoduna göre
spektrofotometrik olarak 340 nm’de yapıldı. Çalışmanın sonuçlarına göre enzim aktivitesini % 50 inhibe eden ilaç konsantrasyon (I50) değerleri kalsiyum glukonat için 19.4 mM,
sodyum bikarbonat için 160 mM oldu. Buna karşın potasyum klorürün herhangi bir etkisi görülmemiştir.
In vitro effects of calcium gluconate, potassium chloride and sodium bicarbonate on human erythrocyte glucose 6-phosphate dehydrogenase
Effects of some electrolytes which are frequently used for treatment, on erythrocyte glucose 6-phosphate dehydrogenase enzyme (G6PD) activity were investigated in in
vitro conditions. G6PD was purified by affinity chromatography. Enzyme activity was determined according to the Beutler method by using a spectrophotometer at 340 nm. According to the results of study. 50% inhibatör drug concertration values (I50) were 19.4 mM for calcium glukonat and 160 mM for sodium bikarbonat in in vitro study. There
was not observed any inhibitör effect of potassium chloride on glucose 6-phosphate dehydrogenase activity. The result of this study revealed that calcium gluconate and sodium bicarbonate but not potassium chloride have significiant inhibititory effect on the activity of G6PD enzyme in in vitro.
