Aim:
Brucellosis is a zoonotic infection that is common in both animals and humans
all over the world, caused by Brucella bacteria. In this study, it was aimed to
identify species of Brucella bacterial isolates isolated in Adıyaman province
and to determine biotype distribution and profiles.
Material and Methods: Fourty patients (22 males, 18
females) that Brucella grew in their
blood cultures,
were included in the study. Patient
samples were incubated for 10 days for blood culture. Positive signaling
samples were plated on 5% sheep blood agar medium. Then, they were incubated
for 48 hours at 37 °C. For those who were identified as Gram negative
coccobacilli, using by Gram staining, serologic confirmation was performed. The
bacterial strains were identified using an antibiogram device. For the
genotyping, the Polymerase Chain Reaction method was used.
Results: Of the
40 patients included in the study, 22 were male and 18 were female. All of the
isolated bacteria were detected as B.
melitensis with fully automated culture antibiogram device. The biovar
studying failed due to an unknown inhibition in the amplification of the
nucleic acid material.
Conclusion: As in our country, B. melitensis was detected as the causative factor in the majority
of cases of brucellosis in humans, in our province too.
Amaç: Brucellosis, Brucella cinsi bakterilerin neden olduğu tüm
dünyada hem hayvanlarda hem de insanlarda yaygın olarak görülen zoonotik bir
enfeksiyondur. Bu çalışmada, Adıyaman ilinde izole edilen Brucella cinsi
bakterilerin türlerinin identifikasyonu yapılarak, biyotip dağılımı ve
profillerinin belirlenmesi amaçlandı.
Bulgular: Çalışmaya
alınan 40 hastanın 22’si erkek, 18’i kadındı. İzole edilen bakterilerin tümü,
tam otomatize kültür antibiyogram cihazı ile
Brucella melitensis (B. melitensis) olarak saptandı. Biyovar tiplendirme
çalışması, nedeni anlaşılamayan bir inhibisyondan dolayı, nükleik materyal
amplifikasyonu başarısızlıkla sonuçlandı.
Sonuçlar: Ülkemizde olduğu gibi ilimizde de insanlarda Bruselloz
olgularının büyük çoğunluğunda etken olarak Brucella
melitensis saptandı.
