Dose oxidative stress increase during short term sleep deprivation in children patients ?

Abstract

Aim: Oxidative stress is defined as the imbalance in the detoxification of antioxidants and reactive oxygen products produced during cellular metabolism. The deficiency of antioxidant leads to errors in oxidative hemostasis. Reactive oxygen products make damage to the macromolecules of the cell like lipid, protein and DNA and this leads to cellular death. Oxidative stress plays role in the pathogenesis of many chronic diseases. We aimed to investigate the relationship between short period sleep deprivation and thiol disulfide parameters of oxidative stress.

Material and Method: Children between 6 months and 6 years were included in our study. Children with high fever, upper respiratory tract infection, diarrhea, vomiting, or who had any chronic infection or who use any drugs were excluded from the study. Children who were sleep deprived for more than 4 hours are assigned as the study group. Children who were not sleep deprived are assigned as the control group. Serum thiol and disulfide were calculated using the colorimetric method of Erel’s.

Results: Ninety five patients were included in the study. Fifty six  (58.9%) were male and 39 (41.1%) were females. The median (IQR) age was 2.0 (2.0) year, the median (IQR) weight was 13.0 (7.0) kg. The median (IQR) period of sleeplessness was 8.0 (4.0) hours. There were no significant differences between the two groups according to serum levels of native thiol, total thiol, disülfide and disulfide/native thiol and disulfide/ total thiol and native thiol/ total thiol ratios. There was mild correlation between serum disulfide level and the sleep deprivation periods, but no statistical correlation between serum thiol levels and sleep deprivation periods.







Conclusion: We conclude that the oxidative stres hemostasis was not affected by sleeplessnes in pediatric pateints.

Abstract

Amaç: Oksidatif stres; hücresel metabolizma sırasında oluşan reaktif oksijen
türlerinin artışı ile onları detoksifiye eden, antioksidanların yetersizliği
sonucu oluşan oksidatif dengenin bozulması olarak tanımlanır. Reaktif oksijen
türleri hücre içi lipit, protein ve DNA gibi makromoleküllere hasar
vererek  hücre ölümüne neden olur. Oksidatif
stres, birçok kronik hastalığın patogenezinden sorumludur. Çalışmamızın amacı
kısa süre uykusuz bırakılan çocuk hastaların oksidatif stres parametrelerinden
biri olan serum tiyol-disulfid düzeyini belirleyerek, uykusuzluk süreleri ile
oksidatif stres arasındaki ilişkiyi araştırmaktır.

Gereç veYöntem: Yaşları altı ay ile 6 yaş
arasında değişen, manyetik
rezonans
 görüntüleme

 öncesi kliniğimize başvuran çocuk hastalar
çalışmaya dahil edildi. Yüksek ateş, akut solunum yolu enfeksiyonu, ishal,
kusma şikayetleri olan veya  majör
konjenital anomalisi olan veya kronik hastalığı nedeniyle sürekli ilaç kullanan
hastalar çalışmaya alınmadı. Çekim öncesi en az 4 saat uykusuz bırakılan
hastalar çalışma grubu, uykusunu tam alan hastalar kontrol grubu olarak
belirlendi. Serum t
iyol ve disülfid düzeyleri Erel ve ark.’nın
geliştirdiği
yeni kolorimetrik metot ile bakıldı.

Bulgular: Toplam 95 hasta çalışmaya dahil edildi. Hastaların
56’sı (%58,9) erkek, 39’u (41,1) ise kız çocuklardı. Hastaların ortanca (IQR)
yaşı 2,0 (2,0) yıl, ortanca (IQR) vücut ağırlığı 13,0 (7,0) kg idi. Manyetik rezonans görüntüleme’den
önce ortanca (IQR) uykusuzluk süresi 8,0 (4,0) saat idi. Nativ
tiyol, total tiyol ve disülfid serum düzeyleri uykusuz ve kontrol hastaların arasında
anlamlı istatistiksel farklılık saptanmadı. Disülfid/nativ tiyol, disülfid/total
tiyol ve nativ tiyol/total tiyol gruplar arasında anlamlı bir istatistiksel
farklılık saptanmadı. En az dört saat
uykusuz olanlarda uykusuzluk
süresi ile disulfid düzeyi arasında orta derecede korelasyon saptandı. Uykusuzluk
süresi ile total tiyol arasında 
istatistiksel olarak anlamlı korelasyon saptanmadı.









Sonuç: Kısa süre uykusuz bırakılan çocuklarda oksidatif
stress dengesinin bozulmadığı kanaatindeyiz.
