Ticari Ve Kendi Hazırladığımız Echınococcus Igg Elisa İle Kist Hidatikli Hastalarda Operasyon Öncesi Ve Sonrası Total Igg Yanıtlarının Değerlendirilmesi

Öz

Bu çalışmada insan kistik ekinokokkozis'inin CE serodi agnozu amacıyla radyolojik ve klinik olarak kesin tanısıkonulmuş CE'li 18, diğer helmintler Trichuris trichiura, Enterobius vermicularis, Toxocara canis visceral larvamigrans , Taenia saginata, Schistosoma haematobium ve Ascaris lumbricoides ile infekte toplam 60 ve kontrolgrubu olarak sağlıklı 100 kişiden toplanan serumlarda, ticari E.granulosusELISA ve koyun karaciğer kist sıvıantijeni ile hazırladığımız ELISA protokolü ile total IgG antikorları aranmıştır. Çalışmada esas olarak kendihazırladığımız E . g r a n u l o s u sELISA kitinin standardize edilerek rutin uygulamaya konulması hedeflenmiştir.Ameliyatı takip eden birinci ve üçüncü aylarda IgG değerlerinde yükselme görülmüşken, onikinci ve ondokuzuncuaylarda bu değerlerin operasyon öncesi değerlerinin de altına indiği belirlenmiştir.Ticari ve kendi hazırladığımız ELISA kitlerine ait sonuçların yapılan istatistiksel analizinde Mann-WhitneyU testi anlamlı bir farklılık tespit edilmemiştir p>0.05 . Elde edilen sonuçlar, koyun karaciğer kist sıvısı kullanılarakstandardize edilen ELISA kitinin rutin olarak CE'nin serodiagnozunda kullanılabileceğini ortaya koymaktadır

Evaluation Of Pre-And Postoperative Total Igg Responses In Patients With Cystic Hydatidosis Using Commercial And In-House Echinococcosis Igg Elisa

Abstract

In this study for the serodiagnosis of human Cystic echinococcosis CE 18 sera from radiological-clinicaldiagnosed patients with CE, 60 patients infected with various helminths Trichuris trichiura Enterobius vermicularis,Toxocara canis, Taenia saginata, Shistosoma haematobium, and Ascaris lumbricoides , and 100 sera from healthypersons were investigated for existence of total IgG antibodies, by commercial and in-house E.granulosusELISA kits from sheep. The aim of present study was to standardize in-house E.granulosusELISA kit for routinelaboratory tests. Pre-and postoperative IgG values evaluated with both tests. In 1st-3rdmonths following theoperation increase in IgG values was observed whereas, the decrease was observed in 12thand 19thmonths, whichwere below the preoperative IgG values.There was no significant difference Mann-Whitney U test between commercial and in-house ELISA kits p>0.05 . These data revealed that the in-house ELISA kit which were prepared by using sheep liver cyst fluidantigens could be used in serodiagnosis of CE
