Spesifik Patojen Free Spf Fare Üretme Çalışması

Öz

Günümüzde laboratuvar hayvanları üzerinde yapılan in vivotest sonuçlarının geçerli ve güvenilir kabuledilebilmesi için deney hayvanlarının belli standartta olması gerekmektedir. Bu çalışmada, basit bir izolatörkullanarak konvansiyonel Swiss albinofare soyundan Spesifik Patojen Free fare üretimi için bir çekirdek kolonioluşturulması amaçlanmıştır.Konvansiyonel olarak yetiştirilmekte olan bir Swiss albinofare soyundan 33 adet gebe dişiye histerektomiuygulanarak yavrulu uteruslar alınmış ve patojen mikroorganizmalardan arınmış izolatöre aseptik koşullardaaktarılmıştır. İzolatör içinde uterusdan çıkarılan yavrular, Spesifik Patojen Free C57BL/6 inbred farelerden oluşturulansütanneler tarafından emzirilmiştir. Bu yavrulardan 178 adedi süt kesim çağında izolatörde bırakılarak, bakım vebeslenmeleri izolatör içinde sürdürülmüştür.Sonuç olarak, 4 haftalık dönemde yapılan kontrollerde Swiss albinofarelerin Spesifik Patojen Free özellikkazandıkları ve 10 haftalık dönemde de bu özelliklerini korudukları görülmüştür

Study Of Breeding Specific Pathogen Free Spf Mice

Abstract

Laboratory animals should have certain qualifications in order that tests carried out them give valid andacceptable results. This study aimed to obtain a nucleus colony from conventional Swiss albinomice for SPF miceproduction in a simple isolator. From a conventinally-bred Swiss albinoline 33 pregnant mice were hysterectomized and uteruses containingthe fetuses were aseptically transferred to a pathogen-free isolator. Baby mice were taken out from the uteruses inthe isolator and were suckled by foster mother consisting of Specific Pathogen Free C57BL/6 inbred mice. Onehundred and seventy eight of these baby mice were left in the isolator at weaning age and their feeding and carewere carried out in the isolator.In conclusion, controls at 4th week of age revealed that Swiss albinomice acquired SPF feature and at 10thweek this feature persisted
