Sentetik Piretroid İnsektisit Tetramethrin’in Albino Fareler’de Mus Musculus Serum Proteinleri Üzerine Etkileri

Öz

Amaç: Bu çalışmada, insan ve hayvan vücudu ile bitkiler üzerinde veya çevresinde yaşayan, besin kaynaklarının üretim, depolanma ve tüketimi sırasında besin değerini düşüren ya da zarara uğratan böcek, kemirici, yabani ot, mantar gibi canlı formlarının yıkıcı etkilerini azaltmak için kullanılan kimyasal maddeler olan sentetik pestisitlerden piretroid insektisit tetrametrinin, albino fare Mus musculus ’lerde serum proteinleri albumin, α-globulin, β-globulin ve γ-globulin üzerine etkilerinin belirlenmesi amaçlanmıştır.Yöntem: Tetrametrinin subletal dozları 250 ve 500 mg/kg/gün 4-8 haftalık farelere 13 hafta süreyle ve ağız yoluyla verilmiştir. Deney sonunda, farelerin kalplerinden alınan kan örnekleri Durheim tüplerine konarak, 20-25 oC’de 45 dakika boyunca pıhtılaşmaya bırakıldıktan sonra, soğutmalı santrifüjde +4 oC , 7000-7500 rpm’de ve 15 dakika süreyle santrifüj edilmiştir. Santifüj işleminden sonra elde edilen yaklaşık 20-30 μl serum örneklerindeki serum proteinlerinin analizi selüloz asetat elektroforez yöntemi kullanılarak gerçekleştirilmiştir.Bulgular: Çalışma sonunda, kontrol grubu, aseton grubu ve tetrametrinin uygulandığı T1 ve T2 gruplarının serumlarındaki albumin, α-globulin, β-globulin ve γ-globulin düzeyi bakımından dişi ve erkek bireyler arasında istatistiksel açıdan önemli bir fark bulunmadığından p>0.05 gruplar arası karşılaştırmalarda cinsiyet farkı göz önüne alınmamıştır. Aseton uygulanan farelerin serum protein sonuçlarının da kontrol grubundan istatistiksel olarak farklı olmadığı tespit edilmiştir p>0.05 . Tetrametrin uygulanan grupların 250 ve 500 mg tetrametrin/kg/gün sonuçları, kontrol grubuyla karşılaştırıldığında; albumin, α-globulin ve β-globulin miktarlarında meydana gelen değişikliğin istatiksel açıdan önemsiz; buna karşın her iki grupta da γ-globulin miktarında, kontrol grubuna göre meydana gelen azalmanın istatiksel açıdan önemli olduğu belirlenmiştir. The Effects of Synthetic Pyrethroid Insecticide Tetramethrin on the Serum Proteins of Albino Mice Mus musculus

Abstract

Objective: In this study, it was aimed to determine the effects of the synthetic pyrethroid insecticide tetramethrin, which is one of the chemicals used to minimize the negative effects decrease of nutritional value during production, storage and consumption of food sources of insects, rodents, wild herbs and fungus living on and/or around human and animal body and plants, on serum proteins albumin, α-globulin, β-globulin and γ-globulin of albino mice Mus musculus .Method: Sub lethal dosages 250 and 500 mg/kg/day of tetramethrin were given orally to mice of 4-8-weeks, for a period of 13 weeks. At the end of the study, blood was collected by heart puncture, and then put into Durheim tubes to coagulate at 20-25oC for 45 minutes. Tubes were centrifuged +4 oC at 7000-7500 rpm for 15 minutes, after which approximately 20-30 µl serum was collected. The analysis of the serum proteins was established by cellulose acetate electrophoresis assay.Results: At the end of the study, gender gap analysis was not taken into consideration between male and female mice, since there were no statistically significant difference p> 0.05 between the levels of the albumin, α-globulin, β-globulin in the serum of the control group, acetone group and tetramethrin applied T1 and T2 groups. It was determined that the results of serum proteins of mice in the acetone applied group were not statistically different from the control group p> 0.05 . Changes of the amounts of the serum albumin, α-globulin and β-globulin in the tetrametrin applied groups 250 and 500 mg tetramethrin / kg / day compared to the control group were statistically insignificant, whereas the results of the γ-globulin were statistically significant in both groups. The results of albumin, α-globulin and β-globulin quantities are compared between T1 and T2 groups and it were statistically insignificant p> 0.05 . On the other hand it was found that decrease in the amount of γ-globulin was statistically significant in T2 group compared to the T1 group results p
