Mikrobiyolojide Kullanılan Hızlı Tanı Yöntemleri

Öz

Mikrobiyoloji uygulamalarında önemli bir yere sahip olan hızlı yöntemler klinik, gıda ve çevresel örneklerde bulunan patojen mikroorganizmalar ve onların metabolitlerinin erken tespiti, izolasyonu, identifikasyonu ve sayımı için yaygın bir şekilde kullanılmaktadır. İlk olarak 1960’ların ortalarında geliştirilmeye başlayan bu yöntemler, minyatürize biyokimyasal, immunolojik, genetik ve biyosensör tekniklerini kapsamaktadır. Minyatürize biyokimyasal identifikasyon yöntemleri 1965 ile 1975 yılları arasında geliştirilmiş, 1975 ile 1985 yılları arasında kalan süre immunolojik test yöntemlerinin altın çağı olarak adlandırılmıştır. Genetik tanı yöntemleri ile polimeraz zincir reaksiyonu PZR uygulamaları 1985 ile 1995 yılları arasında gelişim göstermiştir. 1995 yılından günümüze kadar ise biyosensör ve mikroarray test sistemleri, çeşitli örneklerde bulunan patojen organizmaların direk tanısı amacıyla geliştirilmiştir. Bu derlemede anlatılan immunolojik testler, manüel ve otomatik ELISA Enzyme Linked İmmunosorbant Assay testlerini, lateral migrasyon immunoassay, lateks aglütinasyon testlerini ve immunomanyetik separasyon yöntemlerini kapsamaktadır. Genetik tanı yöntemlerinden birisi olan PZR, patojen mikroorganizmaların doğru tanısını güçlendirmek için son yıllarda daha da geliştirilmiştir. Hızlı testlerin güvenirliği ile ilgili bir çok araştırmanın sonuçları göstermiştir ki; bu testler ucuz ve hızlıdır ayrıca yüksek özgüllük ve duyarlılığa sahiptirler. Rapid Diagnostic Methods in Microbiology

Abstract

Rapid methods are an important part of applied microbiology and are widely used for isolation, identification, early detection, and enumeration of pathogen microorganisms and their products in clinical, food, and environmental samples. These methods which were comprised as miniaturized microbiological, advanced immunological, genetic, and biosensor techniques were began to be developed in mid-1960s. The miniaturized microbiological diagnostic kits were developed from 1965 to 1975. The years of among 1975- 1985 were called the golden age of immunological tests. The genetic diagnostic methods and polymerase chain reaction PCR applications have developed between the years 1985 and 1995. Since 1995, the biosensor and microarray test systems have been developed for direct detection of pathogen microorganisms in different samples. The immunological tests described in this review include manual and automated enzyme linked immunosorbant assays, lateral migration immunoassays, latex agglutination tests and immuno-magnetic separation assays. PCR process which is one of the genetic procedures has been improved in order to strenghten the diagnosis of pathogen organisms. Numerous studies have been conducted to determine the sensitivity and specificity of these tests. They have determined that the tests are rapid, highly sensitive, specific and inexpensive methods. Theoretical information and results of comparative studies about these methods are described in detail in this review.
