ABİETİK ASİT VE GUAİAZULEN TERPENLERİNİN “SEKONDER METABOLİT” GENOTOKSİK POTANSİYELLERİNİN İN VİTRO MİKRONÜKLEUS TESTİ İLE BELİRLENMESİ

Abstract
Bitkiler tarafından sentezlenen terpenler, lezzet, koku ve renk verici maddeler olarak gıda ve kozmetik sektörlerinde, antimutajen, antioksidan ve antikanserojen aktiviteleri ile de tıp ve eczacılık alanında sıklıkla kullanılmaktadır. Farklı alanlarda kullanılan bu bileşikler, insanlar tarafından birçok ürünle birlikte temas veya oral yolla vücuda alınmaktadır. Bu çalışmada doğrudan ya da dolaylı olarak alınan guaiazulen ve abietik asit terpenlerinin doz-süre etkileşimine bağlı olarak genotoksik etkileri belirlenmiştir. Bu amaçla insan periferik kan kültürü kullanılmış ve in vitro mikronükleus (MN) testi ile çalışılmıştır. Terpenlerin farklı konsantrasyonlarını içeren (25,50,100,200ppm) uygulama gruplarına ait veriler terpenlerin çözücüsü olan DMSO kontrol grubu ile karşılaştırılarak istatistiki analizleri yapılmıştır. Ancak guaiazulen terpeninin en yüksek uygulama grubu olan 200 ppm’de hücre proliferasyonu gerçekleşmemiştir. Bu nedenle bu uygulama grubu değerlendirmeye alınamamıştır. Elde edilen verilere göre, guaiazulen ve abietik asit terpenlerinin insan lenfosit hücrelerinde mikronükleus frekansında artışa neden oldukları ve konsantrasyon artışına bağlı olarak sitotoksik ve genotoksik aktivite sergiledikleri belirlenmiştir.
