Abstract

Araştırmada, genç erişkin dönemde kastrasyon yapılan ratların böbrek dokusunda meydana gelen histolojik ve morfometrik farklılıkların incelenmesi amaçlandı. Materyal olarak 2 aylık 36 adet erkek Sprague-Dawley rat kullanıldı. Ratlar 2 kontrol (n=16) ve 2 deney (n=20) olmak üzere toplam 4 guruba ayrıldı. Deney gurubundaki ratlara kastrasyon işlemi yapıldı. Kontrol gurubundaki ratlara ise herhangi bir işlem yapılmadan eşit ortam koşullarında tutuldu. Kastrasyondan 1 ile 2 ay sonraki dönemlerde hem kontrol hem de deney guruplarından doku örnekleri alındı. Histometrik değişimleri belirlemek amacıyla Periodic Acid Schiff (PAS) boyama metodu uygulandı. Işık mikroskobu (Leica DMLB) ve buna bağlı Görüntü analiz sistemi (Leica Q Win Standart) ile morfometrik ölçümler yapılarak (birim alanda korpuskulum renis ve tubulus sayıları ile korpuskulum renis, proksimal tubul, henle kulpu ve toplayıcı borucuk çapları) böbrek histolojisinde meydana gelen değişimler incelendi. Yapılan çalışmada, kastrasyondan hem 1 ay hem de 2 ay sonraki dönemlerde, proksimal tubulleri oluşturan hücrelerin sitoplazmalarında kontrol gruplarına göre PAS pozitif sitoplazmik granüllerin çok yoğun olduğu gözlendi. Morfometrik ölçümler sonucu ise, kastrasyondan 1 ay sonra alınan örneklerde birim alandaki korpuskulum renis sayısı kontrollere göre artarken, korpuskulum renis, henle kulpu ve toplayıcı borucuk çaplarının azaldığı saptandı. Kastrasyondan 2 ay sonra yapılan ölçümlerde, korpuskulum renis ve toplayıcı borucuk çapları artarken, tubulus proksimalis ve henle kulpu çaplarının azaldığı görüldü. Sonuç olarak, kastrasyonun böbrek histokimyası ve morfometrisine yönelik oluşturduğu değişimler ortaya konmuştur
