Acanthus spinosus L.: In Vitro Antimicrobial Activity, Fixed Oil Amount, Mineral Element Analysis, and Fatty Acid Composition (Kireç Plateau/ Kahramanmaraş)

Abstract

In this study, samples of Acanthus spinosus L. were collected from Kireç Plateau (Kahramanmaraş) at 2024. The purpose of the study was to research the antimicrobial activity, fixed oil content, mineral element combination, and fatty acid contents of Acanthus spinosus L. The antimicrobial activity of Acanthus spinosus L. was tested against 12 bacterial species and 1 fungal species using aqueous, methanol, ethanol, and ethyl acetate extracts by the disk diffusion method. The results showed that the ethyl acetate extract exhibited only very slight antifungal activity (7 mm inhibition zone) against Candida albicans ATCC 14053. The fixed oil amount of this plant was showed as 1.80% using the Randall extraction method. The fatty acid contents were analyzed by preparing methyl esters and using GC with a FID detector. The analysis identified 25 different phytochemical fatty acid components, of which 14 were unsaturated (38.452 %) and 11 were saturated fatty acids (61.549%). Capric acid had the highest proportion (31.290%) in the saturated fatty acids, while Oleic acid was the dominant in the unsaturated fatty acid (11.208%). For mineral element analysis, a microwave digestion system and an ICP-OES device were used. The above-ground parts of the plant were analyzed for mineral elements (Na, K, Ca, P, Mg, Fe, Mn, Zn, Cu, Mo). The findings indicate that Acanthus spinosus L. is a potential natural source with a rich fatty acid content and high calcium and zinc content, despite its very limited antifungal source.
