Effect of Using Potassium Acetate Instated of Phenol-Chloroform for DNA Isolation on DNA Yield and Quality

Abstract

The objective of this study was to investigate the effect of using potassium acetate instead of phenol-chloroform on the yield and quality of DNA used for polymerase chain reaction (PCR) assays. For this purpose 13 blood samples collected from Kilis goats were used. After proteinase K digestion, phenol-chloroform extraction or treatment with potassium acetate was performed on aliquots of each sample. Concentration and purity of DNA samples were measured at 260 nm and 260nm/280nm by using a spectrophotometer, respectively. Concentration of DNA in phenol-chloroform and potassium acetate groups were found to be 0.4185±0.2368 and 0.2753±0.0846 respectively. Absorbance ratio values of phenol-chloroform and potassium acetate groups were 1.6380±0.1817 and 1.7435±0.0659 respectively. Differences between groups were statistically significant. The results suggested that sufficient amount of DNA were obtained by using potassium acetate in order to remove proteins. Therefore potassium acetate can be used instead of phenol-chloroform which is more hazardous for the health of researchers or for the environment, although DNA yield obtained by using potassium acetate was lower than that obtained by using phenol-chloroform.
