Hepatoprotective and Antioxidant Properties of Pressure-Blanched Curcuma xanthorrhiza Roxb. Powder in CCl4-treated HepG2 Cell

Abstract

Introduction: Oxidative stress plays a key role in liver diseases such as CCl4-induced injury, highlighting the therapeutic potential of natural antioxidants. Curcuma xanthorrhiza Roxb. (Javanese turmeric), rich in curcuminoids and xanthorrhizol, possesses antioxidant properties. Processing affects bioactive compounds; blanching can preserve them by inactivating enzymes. This study evaluated the antioxidant and hepatoprotective effects of pressure-blanched C. xanthorrhiza powder, processed via different drying methods, against CCl4-induced injury in HepG2 cells. Methods: C. xanthorrhiza rhizomes were pressure-blanched (120 °C/28.81 psia, 0-10 min), then dried (cabinet at 50-55 °C or sun). Powdered ethanol extracts were assessed for antioxidant activity (DPPH, FRAP), total phenolics, flavonoids, curcuminoids, and tannins. Hepatoprotective effect was measured in CCl4-treated HepG2 cells via MTS assay (viability) and AST activity. Results: Both drying methods yielded powder extracts with significant antioxidant activity. Sun-drying showed higher DPPH scavenging; FRAP was optimal after 5 min of blanching for both. Bioactive compound levels varied based on blanching time and drying. Extracts protected HepG2 cells from CCl4 toxicity, improving viability and reducing AST. Discussion: Pressure blanching (especially 5 min) and appropriate drying effectively preserved and enhanced C. xanthorrhiza's antioxidant and hepatoprotective properties. The protection against CCl4-induced injury in HepG2 cells is likely due to high antioxidant capacity and preserved bioactive compounds. These findings highlight pressure-blanched C. xanthorrhiza's potential for liver health, warranting in vivo studies.
