Morphological Identification of Golden Cyst Nematode (Globodera rostochiensis) Populations in Turkey

Abstract

Potato cyst nematodes (PCN) Golden cyst nematode, Globodera rostochiensis (Wollenweber, 1923) Skarbilovich, 1959 and Pale cyst nematode, G. pallida Stone, 1973, are sibling species that cause significant yield losses in potatoes almost all over the world. The accurate and rapid diagnosis of PCN, which are important quarantine pests, is very important for management program. Due to the risks posed by these two species, national survey program is implementing in certificate and table potato producing areas in term of the presence of PCN in Turkey. Within the scope of the study, cysts and second-stage larvae of PCN populations obtained from 10 different locations belonging to Niğde, Nevşehir, İzmir, Afyon, and Bolu provinces were identified according to their morphological and morphometric characters. The study includes the first detailed morphological and morphometric data of G. rostochiensis populations in other provinces except İzmir. As a result of the study, all populations were identified as G. rostochiensis. The results of this study conducted for the first time to cover all of the areas infested with PCN in Turkey were show that measurements values is consistent with literature references and G. rostochiensis populations of our country can be identified morphologically. However, it is thought that it would be more accurate to use morphological and molecular methods together in the diagnosis of PCN, which is one of the most important quarantine pest in routine analysis.

Türkiye'deki Altın Kist Nematodu (Globodera rostochiensis) Popülasyonlarının Morfolojik Tanımlanması

Abstract

Patates kist nematodları (PKN) Altın kist nematodu olarak isimlendirilen Globodera rostochiensis (Wollenweber, 1923) Skarbilovich, 1959 ve Beyaz kist nematodu olarak isimlendirilen G. pallida Stone, 1973, tüm dünyada hemen hemen tüm patates alanlarında önemli verim kayıplarına neden olan yaygın türlerdir. Önemli karantina etmenleri olan PKN’nın doğru ve hızlı teşhisi mücadelesi açısından çok önemlidir. Bu iki türün oluşturduğu risk nedeniyle, Türkiye'de bu etmenlerin varlığı sertifikalı ve yemeklik patates alanlarında ulusal survey programı kapsamında taranmaktadır. Çalışma kapsamında Niğde, Nevşehir, İzmir, Afyon ve Bolu illerine ait 10 farklı lokasyondan elde edilen PKN popülasyonlarına ait kist ve ikinci dönem larvalar morfolojik ve morfometrik karakterlerine göre teşhis edilmiştir. Çalışma İzmir ili hariç diğer illerdeki G. rostochiensis popülasyonlarının ilk detaylı morfolojik ve morfometrik verilerini içermektedir. Çalışma sonucunda popülasyonların tamamı G. rostochiensis olarak teşhis edilmiştir. Türkiye’deki bulaşık alanların tamamını kapsayacak şekilde ilk kez yapılan bu çalışmayla ülkemizdeki G. rostochiensis popülasyonlarının morfolojik olarak teşhisinin yapılabildiği, ölçüm değerlerinin referans literatürlerle uyumlu olduğu belirlenmiştir. Ancak rutin analizlerde tüm dünyada en önemli karantina etmenlerinden biri olan Patates kist nematodlarının teşhisinde morfolojik ve moleküler yöntemlerinin bir arada kullanılmasının daha doğru olacağı düşünülmektedir.
