Descriptive Plate-Level Growth Patterns of Adherent Candida on Heat-polymerized Acrylic Resins Pre-exposed to Different Agents: An In Vitro Pilot Study

Abstract

Objectives This pilot study descriptively evaluated plate-level recovery of adherent Candida albicans (C. albicans) and Candida parapsilosis (C. parapsilosis) from heat-polymerized polymethyl methacrylate (PMMA) specimens pre-exposed to selected natural substances and commercial mouthwashes. Precipitate formation in post-immersion solutions was assessed as a secondary exploratory outcome. Material and Methods Thirty-six heat-polymerized PMMA specimens were allocated to 9 groups: sterile distilled water (control), chitosan, propolis, curcuma, ginger, clove, and commercial 3 mouthwashes [GUM (Sunstar Europe SA, Etoy, Switzerland), Parodontax (Haleon, London, UK), GennaDent (Vivanatura, London, UK)]. After 48 h of pre-exposure at 37 °C, two specimens from each group were assigned to each Candida species and processed together, generating one pooled microbial suspension per treatment–species combination. Following 24 h of fungal incubation, adherent microorganisms were detached by rinsing and vortexing in sterile saline. Undiluted and 10⁻¹ aliquots were cultured on Sabouraud agar, and raw plate-level outputs were recorded using an automated colony counter. Outputs exceeding 300 colonies or originating from dense or partially confluent growth were retained descriptively but were not interpreted as precise quantitative counts. No inferential analysis was carried out. Results Parodontax yielded outputs of 29 (undiluted) and 7 (diluted) for C. albicans and 0 (undiluted) and 0 (diluted) for C. parapsilosis. The corresponding distilled-water control outputs were 1100 (undiluted) and 223 (diluted), and 436 (undiluted) and 37 (diluted), respectively. Fifteen outputs of 36 plate exceeded 300 colonies, precluding reliable quantitative ranking. Precipitates were observed with chitosan, curcuma, ginger, and GUM. Conclusion Natural substances and commercial mouthwashes can exhibit varying levels of antifungal activity on acrylic resins used in denture bases. But the findings represent preliminary descriptive observations and do not establish detailed comparative antifungal efficacy, dilution proportionality, PMMA degradation, or material compatibility.
