In Vitro Hybrid Plant Regeneration by Embryo Rescue in Interspecific Hybridization between Some Cucumis Species

Abstract

In this study, the possibility of gene transferring by interspecific hybridization between C. melo and C. sativus, and also the usage of C. melo var. flexuosus as a bridge between the former two species were investigated. Reciprocal crosses were made among C. melo, C. sativus and C. melo var. flexuosus. Two weeks after controlled pollinations, fruits were harvested and surface sterilized by 30 % hypochlorite. The embryos were rescued from the seeds and in vitro cultured on MS solid media. The percentages of fruit settings were 50 % in C. flexuosus x C. sativus; 46.6 % in C. flexuosus x C. melo; 53.3 % in C. melo x C. flexuosus; 33.3 % in C. melo x C. sativus; 53.3 % in C. sativus x C. flexuosus; 43.3 % in C. sativus x C . melo. The seed number per fruit was lowest in C. flexuosus x C. sativus (117.0) and highest in C. melo x C. sativus (432.8). However, many of the seeds in the combination of C. melo x C. sativus were empty and the percentage of seeds with embryo was lowest (32.2 %). In the combination of C. melo and C. flexuosus, most of the seeds (over 90 %) contained embryos. The plant regeneration rate ranged from 12.5 to % 25.0. A total of 32 plantlets was obtained in vitro from these embryos.

Bazı Cucumis Türleri Arasındaki Melezlemelerde Embriyo Kurtarma Yoluyla InVitro Hibrit Bitki Regenerasyonu

Abstract

Bu çalışmada Cucumis cinsi içinde yer alan C. melo ve C. sativus arasında türler arası melezleme ile gen aktarımını sağlayabilme ve C. melo var. flexuosus’u iki tür arasında köprü türü olarak kullanabilme olanakları araştırılmıştır. Bu amaçla C. melo, C. sativus ve C. melo var. flexuosus arasında melezlemeler yapılmıştır. Kontrollu tozlamalardan iki hafta sonra hasat edilerek laboratuvara getirilen ve % 30'luk hipoklorit çözeltisinde yüzey dezenfeksiyonuna tabii tutulan meyvelerdeki tohumlar açılarak içerisindeki embriyolar çıkarılıp MS ortamında in vitro kültüre alınmıştır. Melezlemelerdeki meyve tutum oranları C. flexuosus x C. sativus tozlanmasında % 50, C. flexuosus x C. melo’da % 46.6, C. melo x C. flexuosus’da % 53.3 ve C . melo x C. sativus’da % 33.3, C. sativus x C. flexuosus’da % 53.3, C. sativus x C. melo’da % 43.3 olmuştur. Meyve başına tohum sayısı ortalama 117 adet ile C. flexuosus x C. sativus kombinasyonunda en düşük, 432.8 ile C. melo x C. sativus’da en yüksek bulunmuştur. Ancak C. melo x C. sativus’daki tohumların çoğunun boş olduğu ve embriyo içeren tohum oranının % 32.2 ile en düşük düzeyde kaldığı görülmüştür. Buna karşın C. melo ile C. flexuosus arasında yapılan melezlemelerde tohumların tamamına yakınının (%90) embriyo içerdiği belirlenmiştir. Embriyoların bitkiye dönüşüm oranı % 12.5 ile % 25.0 arasında gerçekleşmiştir. Bu embriyolardan in vitro kültürde toplam 32 adet bitki gelişmiştir.
