An Investigation on Gene Regions Related to Some Economic Traits in Holstein and Brown Swiss Cattle Breeds by Using PCR-RFLP Technique

Abstract

In this study, the polymorphisms in calpain (CAPN1), estrogen receptor α (ERα), prolactin (PRL), and myostatin (MSTN) genes thought to be related to some economical traits were investigated in 136 heads from Holstein and Brown Swiss cattle breeds reared in Karacabey town of Bursa by using PCR-RFLP method. While MSTN locus was found as monomorphic, 2 alleles of each CAPN1, ERα and PRL genes were determined in both breeds. While 3 genotypes (CC, CT, and TT) were determined at CAPN1 locus of both breeds, 2 genotypes (AG and GG) and 3 genotypes (AA, AG, and GG) Juszczukwere Determined at PRL locus of Holstein and Brown Swiss breeds, respectively. Two genotypes (AG and GG) were detected at ERα locus in both breeds. Both two populations were found at Hardy-Weinberg equilibrium except for PRL locus in Brown Swiss population.

Esmer ve Siyah Alaca Irkı Sığırlarda Ekonomik Özelliklerle İlişkili Bazı Gen Bölgelerinin PCR-RFLP ile İncelenmesi

Abstract

Bu çalışmada Bursa Karacabey ilçesindeki yetiştirilen Siyah Alaca ve Esmer sığır ırkından toplam 136 hayvan çeşitli verim özellikleri ile ilişkili oldukları düşünülen CAPN1, ERα1, PRL ve MSTN genlerindeki çeşitli mutasyonlar bakımından PCR-RFLP yöntemi kullanılarak incelenmiştir.

Myostatin geni bakımından her iki populasyon da monomorfik bulunurken, diğer üç lokus bakımından her iki populasyonda da iki allel belirlenmiştir. İncelenen iki ırkta da CAPN1 lokusunda üç genotip (CC, CT ve TT), PRL lokusunda Siyah Alaca ırkında ve Esmer ırkında sırasıyla iki (AG ve GG) ve üç genotip (AA, AG ve GG) bulunmuştur. ERα bakımından incelenen iki populasyonda da iki genotip (AG ve GG) belirlenmiştir. Esmer ırka ait populasyondaki PRL lokusu dışında her iki populasyonun incelenen lokuslar bakımından HW dengesinde olduğu belirlenmiştir.
