Caryologic Study and Caryotypic Analysis of Two Species Artemisia fragrans Willd., A. absinthium L. Belonging to Genus Artemisia

Abstract

Chromosome study and caryotype analysis of two species A. fragrans Willd., A. absinthium L. belonging to family Asteraceae, genus Artemisia was carried out after collecting specimens belonging to these species from different parts of Arazbaran region; Eastern Azerbayjan province of İran. Root tips collected from roots of germinated seedling were treated with α-bromonaftalein and hydroxy quinolin and were fixed with formaldehyde and acid chromic solution; there after, they were stained with aseto iron hematoxilin. These were used for caryotype analysis at pre metaphase stage. The results showed that A. absinthium L. contained 2n=2x=18 chromosomes and was diploid. A. fragrans Willd. contained 2n=4x=36 chromosomes and was tetraploid. Satellite chromosomes in both species were observed such that paired Satellite was recorded in A. absinthium L. and four paired Satellite were recorded in A. fragrans Willd. Moreover, aneuploid were also noted in both species. Average chromosome length of A. absinthium L. was 6.249 ± 0.126 mµ and in tetraploid species A. fragrans Willd. average chromosome length was 5.010 ± 0.056 mµ

Artemisia cinsinin iki türünün Artemisia fragrans Willd., A.absinthium L. karyolojik incelenmesi ve karyotip analizi

Abstract

Artemisia cinsinin, Asteraceae familyasından iki ayrı tür Artemisia fragrans Willd., A. absinthium L. kromozomlarının incelenmesi ve karyotip analizi için, İran doğu Azerbaycan ilinin Arazbaran Karadağ Bölgesinin farklı ekolojilerinden toplanıp tanımlanmaları yapılmıştır. Belirtilen bu türlerin çimlenen tohumlarından elde edilen kök ucuları α-bromonaftalin ve hidroxy kinolin de ilk işlem yapılıp formaldehit ve kromik asit çözücülerinde tespit edilmiştir. Daha sonra asetik asit ve hematoxilinde boyanmıştır. Mitozun metafaz devresinde karyotip analizi yapılmıştır. Elde edilen sonuç bilgilere göre; A. absinthium L. türü 2n=2x=18 kromozom içermekte ve diploid, A. fragrans Willd. türü ise 2n=4x=36 kromozom içermekte ve tetraploid olduğu anlaşılmıştır. Her iki türde de satelitli kromozomlar görülmüş ve diploid A. absinthium L. bir çift satelit, A. fragrans Willd. ise tetraploid ve dört çift satelit içeren kromozomlara sahiptir. Ayrıca her iki türde de aneuploidi ile karşılaşılmıştır. Kromozomların ortalama boyu, diploid tür A. absinthium’da 6.249 ± 0.126 mµ ve tetraploid tür A. fragrans’da ise 5.010 ± 0.056 mµ olarak ölçülmüştür
