The Use of Spectrophotometric Method to Determine Solanapyrones Production of Ascochyta rabiei Pass Labr., Causal Agent of Chickpea Blight

Abstract

This study is aimed to determine solanapyrones production of Ascochyta rabiei by spectrophotometric method and to define the most suitable day for solanapyrones extraction. Three isolates of A. rabiei were grown in czapek dox liquid medium for 7, 14 and 21 days and toxins were extracted by solid phase extraction using C18 catridge. Then toxin were eluted with acetonitrile. Agilent Specord 200 spectrophotometer was used in quantities analysis of solanapyrones. Toxin production of the isolates on 7th, 14th and 21th days were found diffrent from quantitatively. All three isolates produced solanapyrone B on the 7th day. Two of the isolates, except Auk 7, produced solanapyrone A in very low quantities in the 7th day. Amount of solanapyrone A obtained from 14th day was higher than that of 7th day incubation. This implies as that the increase of mycelial mass of fungi is closely related with quantitive of solanapyrone A. In the 7th, 14th and 21th days extraction, solanapyrone C was defined the most abundant toxin for all isolates. The differencies amoung of the quantities solanapyrone A, B and C was not statistically significant between in the 14th and 21th days of incubation. Consequently it is found that 14th day’s incubation appropriate for toxin extraction of A. rabiei

Nohut yanıklık etmeni Ascochyta rabiei Pass Labr.’nin Solanapyrone üretiminin saptanmasında spektrofotometrik yöntemin kullanılması

Abstract

Bu çalışmada Ascochyta rabiei’nin solanapyrone toxin üretiminin spektrofotometrik yöntemle tespiti ve toksin üretiminde en uygun günün belirlenmesini amaçlanmıştır. A. rabiei’nin üç izolatı Abk 3, Auk 7 ve Haypa 3 Czapek Dox sıvı besin ortamında 7, 14 ve 21 gün süreyle geliştirilmiş ve toxin C18 katı faz ekstraksiyon kartijleriyle ekstrakte edilerek acetonitril ile elute edilmiştir. Solanapyrone toksinlerinin kantitatif miktarlarının belirlenmesinde Agilent Specord 200 spektrofotometre kullanılmıştır. A. rabiei izolatlarının 7, 14 ve 21. günlerdeki solanapyrone A, B ve C üretim durumlarının birbirinden farklı olduğu görülmüştür. Üç izolatın da 7. gün ekstraksiyonlarında solanapyrone B ürettiği belirlenmiştir. Solanapyrone A’yı izolatlardan ikisinin düşük miktarda da olsa ürettiği ancak Auk 7 izolatının üretmediği tespit edilmiştir. 14. gündeki solanapyrone A miktarları 7. gündeki miktarlardan oldukça yüksektir. Bu da misel kitlesindeki artışı ile solanapyrone A miktarı arasında ilişki olduğunu göstermektedir. Ekstraksiyon yapılan günlerde her 3 izolatın da en çok solanapyrone C ürettiği belirlenmiştir. İstatiksel olarak bir kıyaslama yapıldığında üç izolatın solanapyrone A, B ve C üretimlerinin 14. ve 21. günler arasında bir fark olmadığı görülmüştür. Sonuç olarak A. rabiei’nin toksin ektraksiyonu için en uygun günün 14. gün olduğu saptanmıştır
