RECOVERY OF ENTEROCOCCUS FAECALIS FROM DIFFERENT TYPES OF TURKISH CHEESE

Abstract

Aim: Food-borne viable Enterococcus faecalis cells contaminate the root canal during treatment due to inadequate aseptic control or penetrate the root filling via coronal leakage after root canal treatment. Enterococci are commonly found in cheese, meat products, olives and vegetables. The aim of this study was to investigate E.faecalis in 18 different cheese in Cukurova region. Materials and method: Eighteen different types of cheese were provided from local supermarkets and investigated for their enterococci content. 1-g pieces were cut out from the body of each cheese using sterile instruments. The cheese pieces were homogenized for 1 min in 10 mL of a 2% sodium citrate buffer using a tissue homogenizer. Subsequently, 10-fold serial dilutions were prepared in 2% sodium citrate down to 10-5. As much as 50 μL of each dilution was plated on kanamycin aesculin azide agar (KAAA) using a spiral diluter and incubated at 42 ºC in a 5% carbon dioxide atmosphere. Colonies were counted after 3 days of incubation. Enterococcus colonies were presumptively identified by their blackening of the agar due to hydrolysis of esculin. Colonies that displayed the typical enterococcal growth and cell morphology were picked up and subcultured on KAAA plates. Subsequently, a catalase test was performed and Gram-positive, catalase-negative cocci were identified to species level by Vitek-2 automatized system according to the instructions of the manufacturer. Result: None of the cheese samples contained viable E.faecalis, 3 cheese sample harbored E.faecium, 2 cheese sample harbored E.durans, 1 cheese sample contained E.gallinarum and 2 cheese sample harbored Pediococcus spp. Conclusion: E.faecalis occur in a variety of cheeses, especially in Europe. And they find the relation enterococci in cheese and their occurence in the oral cavity. We couldn’t found E.faecalis in Turkish cheese so it could be the reason of decreasing percentage of E.faecalis in root canals

Abstract

Amaç: Gıda kökenli yaşayabilen Enterococcus faecalis hücreleri, tedavi esnasında yetersiz aseptik kontrolden dolayı kök kanalını kontamine edebilir veya kanal tedavisinden sonra koronal sızıntı yoluyla kök kanal dolgusuna nüfuz edebilir. Enterokoklar yaygın olarak peynir, et ürünleri, zeytin ve sebzeler gibi çiğ tüketilen gıda türleri içinde veya üzerinde bulunur. Bu çalış- manın amacı bölgemizdeki 18 farklı peynir türündeki E.faecalis varlığını incelemektir.

Gereç ve yöntem: Yerel süpermarketlerden temin edilen peynirlerden steril araçlar kullanılarak 1 gramlık parçalar kesildi. Bu peynir parçaları 10 ml %2’lik sod- yum sitrat buffer içerisinde bir dakika süre ile homo- jenize edildi. Daha sonra %2’lik sodyum sitrat buffer ile 10 kat seri dilüsyonları hazırlandı (10-1-10-5). Her bir dilüs- yondan 50 μL kanamisin eskülin azid agara (KAAA) ekim yapıldı ve 42oC’de, %5 CO2’li ortamda inkübe edildi.  Üç günlük inkübasyondan sonra koloniler ince- lendi. İlk olarak eskülin hidrolizi sonucu siyah renkli koloni oluşturan bakteriler enterokok kolonileri olarak tanımlandı. Tipik enterokok koloni görünümü olan ko- loniler KAAA kültür plaklarına subkültüre edildi. Daha sonra Gram boyama ve katalaz testi ile incelendi ve Gram pozitif, katalaz negatif koklar, üreticinin önerile- rine göre Vitek-2 otomatize sistemi ile tür düzeyinde tanımlandı.

Bulgular: Bu peynir numunelerinin hiçbirinde yaşa- yabilen E.faecalis bulunmazken, 3 peynir numunesinde E.faecium, 2 peynir numunesinde E.durans, 1 peynir numunesinde E.gallinarum,  ve 2 peynir numunesinde de Pediococcus spp barındırdığı gözlemlenmiştir.

Sonuç: E.faecalis, özellikle Avrupa’daki pek çok peynir türünde görülmektedir ve bu peynirlerdeki entero- koklar ile bunların oral kavitede bulunması arasındaki ilişkiyi ortaya koyan çalışmalar yapılmıştır. Bu çalışma- da Türk peynirlerinde E.faecalis’e rastlanmamıştır, bu durum kök kanallarında E.faecalis yüzdesini düşüren bir neden olarak sunulabilir.
