Cennet Bambusu (Nandina domestica Thunb.) ‘Seika’ Çeşidinin in vitro Çoğaltımı ve Köklendirilmesi

Abstract

Cennet bambusu adıyla bilinmekte olan Nandina domestica, anavatanı Asya olan, özellikle Japonya, Çin ve Hindistan’a özgü olan ılıman iklimlerde daha iyi yetişen bir bitki türüdür. Yeşilden kırmızıya dönen renkli yapraklarıyla bahçelerde ve peyzaj düzenlemelerinde dekoratif bir öğe olarak kullanılmaya başlanan süs bitkisidir. Gittikçe popüler olmaya başlayan bu bitkinin üretimindeki düşük çoğalma, çelik ile köklenmede karşılaşılan zorluklar nedeniyle doku kültürü avantajlarından yararlanarak çoğaltım ve köklendirme protokolü oluşturulması amaçlanmıştır. Bu amaçla in vitro ortamda steril edilen sürgün parçaları (eksplantlar) WPM ve MS temel besin ortamlarında kültüre alınmıştır. En iyi kardeşlenme oranı MS 1.5 mg/l BAP, 0.1 mg/l NAA, %3 sakaroz, pH:5.7 ortamında elde edilmiştir. En yüksek köklenme ise 1/2MS 0.5 mg/l IBA, 1 mg/l NAA, 0.2 g/l AC, %2 sakaroz, pH:5.7 olan ortamda elde edilmiştir.

In vitro Propagation and Rooting of Heavenly Bamboo (Nandina domestica Thunb.) ‘Seika’ Variety

Abstract

Nandina domestica, also known as heavenly bamboo, is a plant species that grows better in temperate climates, especially in Japan, China and India, and is native to Asia. It is an ornamental plant that has begun to be used as a decorative element in gardens and landscape arrangements with its colorful leaves that turn from green to red. Due to the low reproduction in the production of this increasingly popular plant and the difficulties encountered in rooting with cuttings, it was aimed to establish a reproduction and routing protocol by utilizing the advantages of tissue culture. For this purpose, shoot fragments (explants) sterilized in vitro were cultured in WPM and MS basic nutrient media. The best multiplication was obtained in MS 1.5 mg/l BAP, 0.1 mg/l NAA, 3%sucrose, pH: 5.7. The highest rooting was obtained in 1/2MS 0.5 mg/l IBA, 1 mg/l NAA, 0.2 g/l AC, 2%sucrose, pH:5.7.
