DİABETİK RATLARDA N-ASETİL GLİKOZAMİN EKSPRESYONU VE LOKALİZASYONU

Abstract




AMAÇ: Bu çalışmada diabetik ratlarda diş eti dokusu gingiva da WGA Lektinin lokalizasyonu ve ekspresyonunun araştırılması amaçlandı.

YÖNTEMLER: Toplam 20 adet Wistar dişi sıçan iki gruba ayrıldı ve incelendi. 10 adet sıçan kontrol grubu olarak seçilirken kalan 10 adet sıçan ise diyabetik deney grubu olarak çalışıldı. Yirmi bir günlük deney süresinden sonra sıçanlar dekapitasyon yöntemi ile öldürüldü. Diş eti dokusundan 6 μm inceliğindeki kriostat kesitler alındı. Gruplar N-Asetil Glikozamin’e spesifik (WGA Triticum vulgaris agglutinin) lektini kullanılarak histokimyasal olarak incelendi. BULGULAR: Çalışmamızda WGA kullanılarak yapılan boyamalarda, her iki gruptada boyanmanın gerçekleştiği ve kontrol grubunda boyanma deney grubundakilerine oranla daha fazla olduğu görülmüştür.

SONUÇ: Diş eti kas dokusunda bulunan glikoproteinlerin N asetil glikozamin ünitelerine spesifik lektinle yapılan histokimyasal boyamalar sonucunda diabetli dokularda; kontrollere göre N asetil glikozamin ünitelerine bağlanmanın daha düşük olduğu görüldü.

EXPRESSION AND LOCALIZATION OF N-ACETYL GLUCOSAMINE IN DIABETIC RATS

Abstract




OBJECTIVE: In this study, the WGA lectin gingival gum tissue in diabetic rats to investigate the localization and expression.


METHODS: A total of 20 Wistar female rats were divided into two groups. The remaining 10 rats were selected as a control group of 10 rats in the experimental group were diabetic. Twenty-one days after the experimental period, the rats were killed by decapitation method. Six-μm thick cryostat sections that were obtained from the gingiva of all rats were examined by histochemical staining. Groups were analyzed histochemically using N-Acetyl Glucosamine specific (Triticum vulgaris agglutinin WGA) lectin.


RESULTS: In this study using WGA staining, staining occurred in both groups and the control group was more staining than the experimental group was observed.


CONCLUSION: Glycoproteins in the muscle tissue of the gum as a result of histochemical staining with lectin that specific N acetyl glucosamine units, connecting to N acetyl glucosamine units diabetic tissues were lower compared to the control.
