The genomic DNA isolation methods comparative analysis upon some Sideritis Labiateae and Serratula Asteraceae taxa

Abstract

There are different protocols for DNA isolation from plants. Due to variation in chemical composition of plants, different DNA isolation methods may be necessary for closely related species. In this study, some species belonging to aromatic plant genus Sideritis L. Sideritis vulcanica Hub.-Mor., Sideritis montana L. subsp. montana, Sideritis gulendamiae H. Duman et Karavelioğulları, Sideritis congesta P.H. Davis et Hub.-Mor. and Sideritis tmolea P.H. Davis and some species belonging to the genus Serratula L. Serratula bornmuelleri Azn, Serratula haussknechtii Boiss., Serratula serratuloides DC Takht., and Serratula hakkiarica P.H. Davis were used. The methods used in this investigation included Phenol-chloroform-isoamyl alcohol isolation procedure, CTAB DNA isolation procedure, and procedure from a commercial kit. Purity and the amount of obtained gDNAs were determined via gel electrophoresis and spectrophotometry A260/A280 . Analysis of the results suggested that the CTAB method was the most appropriate for genomic DNA isolation from the taxa in question, to obtain the greatest amount in 1 µl stock solution as the ng amount
