Cytotoxic effects of Satureja cuneifolia extract in liver cancer cell line HepG2

Abstract

In this study, it was aimed to investigate the effects of extracts obtained from the seed of Rock thyme Satureja cuneifolia on the cytotoxic cell death in HepG2 human liver cancer cell lines. The plant samples collected from the vicinity of Kanlıpınar Village of Eskişehir were grounded after drying and then extracts were obtained. The liver cancer cells were replicated in a specially prepared medium. Plant extracts taken at doses of 1, 5, 10, 15, 20, 25 and 30 μg/μl were added to replicated liver cancer cells and the effects levels were examined at 24, 48 and 72 hour intervals. In the study, the data obtained after 24 hours showed that the doses of 1, 5, 10, and 15 μg/μl inhibited the development of cancer cells while 20 μg/μl dose was not effective, the doses of 25 and 30 μg/μl were not effective on the development of cancerous cells. According to the results of 48 hours of experiments; a significant death was observed in cancer cells at all doses applied 1-30 μg/μl . According to the results of 72 hours of experiments; all doses applied 1-30 μg/μl inhibited the development of cancer cells and caused significant death of cancer cells. As a result, it has been determined that Satureja cuneifolia extracts inhibit the development of liver cancer cells and that this plant may be used in the design of cancer medicines. Future studies are needed to determine the most effective dose/time applications and action of mechanisms of Satureja cuneifolia extracts

Karaciğer kanser hücre dizilerinde HepG2 Satureja cuneifolia ekstresinin sitotoksik etkileri

Abstract

Bu çalışmada Kaya kekiği Satureja cuneifolia bitkisinden elde edilen ekstrelerin HepG2 insan karaciğer kanser hücrelerinde in vitro sitotoksik hücre ölümü üzerine etkilerinin araştırılması amaçlanmıştır. Bitki örnekleri Eskişehir’in Kanlıpınar Köyü çevresinden toplanarak kurutulduktan sonra öğütülmüş ve ekstreleri elde edilmiştir. Karaciğer kanser hücreleri özel olarak hazırlanmış olan besi ortamında çoğaltılmıştır. 1, 5, 10, 15, 20, 25 ve 30µg/µl’lik dozlarda alınan bitki ekstreleri, çoğaltılan karaciğer kanser hücrelerine verilmiştir ve etki düzeyleri 24, 48 ve 72 saat aralıklarla incelenmiştir. Kanser hücrelerinde canlılık testleri tripan mavisi ve MTT testi yöntemi kullanılarak araştırılmıştır. Çalışmada 24 saat sonunda elde edilen veriler, 1, 5, 10 ve 15µg/µl dozların kanser hücrelerinin gelişimini engellemesine karşın; 20, 25 ve 30µg/µl dozların ise kanserli hücrelerin gelişimi üzerinde etkili olmadığını göstermiştir. Deneylerin 48 saat sonucuna göre; uygulanan tüm dozlarda 1-30µg/µl kanser hücrelerinde belirgin şekilde bir ölüm gözlenmiştir. Deneylerin 72 saat sonucuna göre; uygulanan tüm dozların 1-30µg/µl kanser hücrelerinin gelişimini engellediği ve kanserli hücrelerin önemli oranda ölümüne neden olduğu belirlenmiştir. Sonuç olarak; Satureja cuneifolia ekstrelerinin karaciğer kanser hücrelerinin gelişimini engellediği ve kanser ilaçlarının tasarımında bu bitkinin kullanılabileceği belirlenmiştir. İleride yapılacak ayrıntılı çalışmalarla Satureja cuneifolia ekstrelerinin en etkin doz/süre uygulamaları ve etki mekanizmalarının belirlenmesi gerekmektedir
