Asma virüslerinin tanılanmasında DAS-ELISA ve RT-PCR yöntemlerinin duyarlılıklarının karşılaştırılması

Abstract



Bu çalışmada asma
virüslerinin tanılanmasında kullanılan farklı yöntemlerin (DAS-ELISA and RT-PCR) duyarlılık durumlarının
karşılaştırılması amaçlanmıştır. Manisa ve İzmir illerinde bağ alanlarında güdümlü
örnekleme yapılmış ve virüs belirtisi gösteren asma bitkilerinden toplam 50
örnek alınmıştır. Bu örneklerdeki viral etmenlerin tanılanmasında DAS-ELISA ve
RT-PCR yöntemleri kullanılmıştır. Total RNA ekstraksiyonu silica temelli metoda
göre sürgünlerin yaprak/ floem dokusundan gerçekleştirilmiştir. Saflaştırılan
RNA revers transkripsiyonda cDNA sentezi için kalıp olarak kullanılmıştır.
Çoğaltılan PCR ürünleri agaroz jel’de yürütülmüş ve ethidium bromide ile
boyanarak gözlemlenmiştir. DAS-ELISA ve RT-PCR testleri sonucu asma
bitkilerinde GFLV, GFkV, ArMV, GLRaV 1,2,3 ve 4-9 etmenleri saptanmıştır. PCR
analiz sonuçları, DAS-ELISA sonuçları ile paralel olmuş ve birbirini
doğrulamıştır.

Comparison of DAS-ELISA and RT-PCR methods for the diagnosis of grapevine viruses

Abstract



The present study was conducted to compare the sensibility of different
methods (DAS-ELISA and RT-PCR) which are used to detect the agents of viral
diseases in grapevine. Purposive sampling was done in vineyards in Manisa and
İzmir provinces and a total of 50 samples were taken from grapevines showing
virus symptoms. DAS-ELISA
and RT-PCR methods were used for identification of viral agents
in these samples. Total RNA extractions were made from serially diluted fresh
leaf tissue, root, bark and one year old green bark tissue using silica-based
method. Purified total RNA extracts were used as template for cDNA synthesis by
reverse transcription. PCR products were analysed by gel electrophoresis and
visualized by ethidium bromide staining. DAS-ELISA test results showed that the
plants were infected with GFkV, ArMV, GLRV 1, 2, 3 and 4-9 viruses. PCR
analysis confirmed DAS-ELISA results.
