Determination of resistance to <em>Bean common mosaıc virus</em> (BCMV) and <em>Bean common mosaic necrosis virus</em> (BCMNV) in some common bean genotypes using qualitative, quantitative and molecular methods
Abstract
In 2007, a total of five bean cultivars, two of which are commercial cultivars, and one breeding line were evaluated for presence of known resistance genes and resistance levels to BCMV and BCMNV using qualitative, quantitative and molecular methods. Five plants of each cultivars and breeding line were inoculated with NL-3 strain of BCMNV and NL-4 strain of BCMV, and all plants were evaluated for virus symptoms and vigor. Additionally, plants were tested by DAS-ELISA to evaluate the virus titer on non-inoculated trifoliate leaves quantitatively. Also, two SCAR markers, SW-13 linked to the dominant I gene and SBD-5 for the recessive gene bc-12 were used to screen these genes in those cultivars by PCR. Results showed that cultivars Zulbiye and Ozeren Seker and 4F-3260 breeding line posses the dominant I gene which is responsible for resistance to BCMV, and Akdag has the recessive gene bc-22 which confers resistance to all known strains of BCMNV and BCMV except NL-4. Cultivars Ozeren Seker and Ozeren Barbunya occurred to possess the recessive gene bc-12 which is responsible for resistance to four strains of BCMV (NL-1, NL-7, US-2, NL-2) and NL-8 strain of BCMNV.
