Research on the Presence of Some Viruses in Olive Orchards Using Serological and Molecular Techniques in Mardin Province of Turkey
Abstract
Surveys were performed in 2010 in order to investigate the presence of several virus diseases in olive trees grown in Mardin province and counties located in the Southeastern Anatolia. Totally 170 shoot samples were collected from trees showing virus symptoms and also collected randomly symptomless trees considering the possibility of virus presence latently. Olive plant samples were tested against Olive latent-1 virus (OLV-1), Olive latent-2 virus (OLV-2), Cherry leaf roll virus (CLRV), Arabis mosaic virus (ArMV), Strawberry latent ringspot virus (SLRSV), Olive latent ringspot virus (OLRSV), Cucumber mosaic virus (CMV) and Olive latent-3 virus (OLV-3). To identify viral agents, the serological (DAS-ELISA) tests using available polyclonal antisera (SLRSV, ArMV, CLRV and CMV) and molecular methods (dsRNA, RT-PCR) were applied to collected olive samples. According to results of DAS-ELISA and PCR analysis, all samples were found negative whereas dsRNA profiles were observed in 41 samples. All tested samples were found free of tested viruses but it was concluded that they have to be tested against other known viruses of olives.
