Morphological characteristic of <em>Cercospora beticola </em>(Sacc.) from infected sugar beet leaves and determination of its growth medium.

Abstract

This study was carried out at the sugar beet field in Oltu (Erzurum) about 30-50% of the area was infected by Cercospora beticola  (Sacc.). Isolates were collected from the infected sugar beet leaves. The collected isolates were transferred into the plates, which contain PDA (Potato Decroze Agar) and SBLA (Sugar Beet Leaves Extract Agar). After the germination, Cercospora beticola (Sacc.) showed differences between conidia and conidiophore sizes. Cercospora beticola sporulated in SBLEA placed on cellophore disc on water agar. Hyphae anastomosis was observed on medium during the extensive growth of hyphae, which formed by germination of conidia placed on cellophore disc on the water agar. Later on it was found that on certain points the growing hyphal produced stromata where conidia and conidiophore were developed.

Enfekteli şeker pancarı yapraklarından izole edilen <em>Cercospora beticola </em>Sacc.'nın morfolojik özellikleri ve besi ortamındaki gelişiminin belirlenmesi.

Abstract

Bu çalışma Erzurum yöresinde, Oltu ilçesi ve civarında şeker pancarı üretimi yapılan alanlarda %30-50 ürün kaybına neden olan Cercospora beticola Sacc. hastalık etmeni üzerine araştırmaları kapsamaktadır. Cercospora beticola ile enfekteli şeker pancarı bitkisinin yapraklarından izolatlar elde edilmiş, bu izolatlar enfekteli yaprak dokusu üzerinden alınarak PDA (Patates Dekroz Agarı) ve SBLEA (Şeker Pancarı Ekstrakt Agarı) besi ortamlarına aktarılmıştır. Besi ortamında kültüre alınan Cercospora beticola hastalık etmeninin konidi ve konidiofor boyutlarında farklılıklar belirlenmiştir. SBLEA ortamında sporlandırılan Cercospora beticola'nın bir kaç konidisi alınarak su agarı üzerine yerleştirilen selefon disklere konulmuş, selefon diskler üzerine konulan konidilerin çimlendikleri, bazı hiflerin anastomosis oluşturdukları ve zamanla bu hiflerin belirli noktalarda üzerinde konidi ve konidiofor oluşacak olan stomaları oluşturdukları saptanmıştır.
